The detection of biomolecules has been an important field in the biomedical engineering subject.Metalloporphyrin(porphyrin)is a conjugated organic metal complex,which can simulate the active centers of many kinds of enzymes.It has a wide range of applications inthe field of catalytic research.In this paper,we combined the click chemistry technology and used porphyrin compounds to catalyze the atom radical polymerization(ATRP)reaction to amplify the electrochemical signal,and applied it to the biosensor of thrombin molecule.1.In this experiment,first,we use two aptamers to form a sandwich structure with thrombin,and the thrombin will be measured on the surface of Au electrode.Through the method of click chemistry,copper ion was reducted into monovalent one and catalyzed azide alkyne radical cycloaddition reaction,which could connect the ATRP initiator to aptamer at the end.Subsequently,the ATRP reaction was introduced into the system,and the porphyrin(Hemin)was used to catalyze the reaction.The ATRP reaction monomer with ferrocene was polymerized into long chains,and the signal amplification was carried out,so that the thrombin could be detected.The experiment shows that the reaction can detect the concentration of thrombin as low as 0.12 fM,and sensitivity has been increased for more than three orders of magnitude compared with the traditional thrombin detection methods.2.In this experiment,we used the same method of the immobilization of thrombin on the gold electrode surface.The monovalent copper ions catalyzed click chemistry was reducted by electrochemical method from copper sulfate.And manganese porphyrin compound was utilized in the ATRP reactions as the catalyst.Thrombin sensor with higher sensitivity was constructed,and the detection limit and linear range of detection were 0.08 fM and 10-14?10-9M respectively.At the same time,the optimizations of the experimental conditions show that,the detection process has the advantages of simple operation,low cost,short time.And it also shows good anti-interference ability in the dilution of serum samples.In summary,this detection method of Thrombin has great potential applications in the field of clinical diagnosis and biology.
