Establishment And Application Of A Mass Spectrometry Based Targeted Metabonnmics

A mass spectrometry analysis method for amino acids,carnitine and some amino metabolites was established.The chromatographic column,mobile phase,gradient mode,column temperature and other parameters were optimized,and the best liquid chromatographic separation method was established.Lipid targeted metabonomics is based on a liquid chromatography coupled with high resolution mass spectrometry platform.Lipids were identified by lipid maps database and verified by secondary mass spectum.Some lipid standard products were also used to verify.Finally,more than 300 species of lipid metabolites were quantified.The method of liquid phase separation is also optimized.The development and full validation of the rapid and sensitive LCMS-based method for determination of amino acid,carnitine and lipid in bio-marices was reported.The accuracy and reliability of the method was verified mainly from the detection limits,quantitative limits,linearity,stability,precision and recovery rate.The results showed that the linear correlation coefficient R2 was greater than 0.999,and the intra day intraday precision was less than 5%.Stability,precision and recovery rate also met the requirements of targeted metabonomics.It was proved that the method was sensitive and accurate,and it can be applied to target group research.In order to verify the applicability of the targeted omics method,a metabonomics analysis of biological matrix was carried out in this study.The selected biological matrix was the plasma of normal people and metabolic syndrome patients.The collected data are processed by data pretreatment and statistics.The res?lts showed that the metabolite detection results had good stability and accuracy.There was a great difference between the normal group and the disease group in amino acids and lipid metabolites.To sum up,LC-MS based platform for different types of metabolites was established,and the application of the practical biological matrix was studied.The target analysis method established in this study covered amino acids,carnitine,lipid more than 400 species of metabolites.These metabolites are involved in many metabolic pathways,which play an important role in the growth and reproduction of organisms,the maintenance of their structure and the response to the external environment.
