Apoptotic bodies are vesicles formed by cell disintegration at the end of apoptosis if apoptotic bodies are incomplete elimination will lead to various diseases even death.In vivo.The Innexins?Inxs?transcriptional level of the hemocyte is significantly reduced,and the apoptotic bodies are formed after Spodoptera litura is parasitized by Microplitis bicoloratus.At the same time,we found that the apoptotic bodies in the medium could be absorbed by cells were overexpressed FoF1-ATPase?.However,the interaction between apoptotic bodies and Inxs and FoF1-ATPase?is not clear.In this study,Sf9 cells were treated with different stimuli,and cells can form apoptotic bodies under the stimulation of Microplitis bicoloratus bracovirus?Mb BV?.In the study of the relationship between apoptotic bodies and the expression of Inxs,it was found that Inxs silent cells could not form apoptotic bodies,but when adding Mb BV after Inxs was silent,the number of cells could form apoptotic bodies was increased,which indicates that the formation of apoptotic bodies was related to the expression of Inxs under the stimulation of Mb BV.Previous studies found that the caspase of the cells was activated after the cells were stimulated by Mb BV.By predicting the caspase cutting site,we discovered that Inxs could be cut by activated caspase,and cause space conformation change of the Inxs,resulting in the formation of apoptotic bodies.At the same time,Sf9 cells were infected by the recombinant baculovirus re Bac-Inx3 which can change the spatial conformation of Inx3,the cells also form apoptotic bodies,indicating that the spatial conformation changes of Inxs will lead to the formation of apoptotic bodies.The study of FoF1-ATPase?was found that FoF1-ATPase?was stable in the cell membrane,and could not be affected by apoptosis factors.The cell's ability to accept apoptotic bodies decreased when FoF1-ATPase?was closed on the cell surface;after overexpressing FoF1-ATPase?,the cells could absorb more apoptotic bodies.Also,the mutation of lysine in Walker A,A conservative structure of FoF1-ATPase?,found that the cell's ability to accept apoptotic cells also decreased.The above results indicated that FoF1-ATPase?was the receptor of apoptotic cells on the surface of the cell,and lysine in the FoF1-ATPase?conservative domain Walker A plays an essential role in the absorption of apoptotic bodies.Further study of FoF1-ATPase?was found that the cells that overexpressed FoF1-ATPase?were more susceptible to apoptosis in vitro.After FoF1-ATPase?was silenced,the apoptosis rate of lymphocyte was significantly decreased,and the growth and development of Spodoptera lituraw were significantly inhibited.To sum up,the alteration of Inxs conformation will lead to the formation of apoptotic bodies,The apoptotic bodies were bound to the FoF1-ATPase?of the cell surface and then were eliminated.
