Anthracnose of maize(Zea mays),caused by Colletotrichum graminicola,is one of the most economically important diseases of maize worldwide?The fungus can infect all plant parts,but the most frequent symptoms are stalk rot and leaf blight,which threaten the Global food security.Thus,it's important to make the pathogenesis clear for preventing the development of Anthracnose of maize.Our lad have got a mutant called ASADH which is an important enzyme from Aspartic acid family,infecting the biosynthesis of amino acids.when The organism cannot synthesize amino acids,we got a kn?kout mutant which called general amino acid permease GAP3,can transport a mino acids in environment to sustain its growth and development.A general amino acid permease GAP3 gene in Magaporthe oryzae was used as a starting point.By sequence alignment we found that,the homology between Colletotrichum graminicola and Magaporthe oryzae was 62.65%.After bioimformatics analysis,GAP3 have transmelain region can transport animosity acid in environment.The biological characteristics and pathogenicity analysis of GAP3 were found that compared with WT,The color of colony of ?GAP3 in CM is all white and in PDA is partial white,while in WT,The color of colony of WT is sepia;Thequantity of sporulation in mutant is lesser than WT;The germination rate of spores is lower than WT and compared the melanin and appressorium with WT,the generation of appressorium is slower and the production of melanin in mutant is decreased.We also found that there are obvious difference between?GAP3 and WT.To the sensitivity to hydrogen peroxide.From the consequence of DAB staining and analysis of hydrogen peroxide concentration,we conclude that general amino acid permease enfect the way to Transfer and absorption of hydrogen peroxide.The relevent stress experiment were performed in article.SDS,Congo-Red,Sorbitol,Na CI,we found that ?GAP3 in SDS and Congo-Red CMMediumare not sensitive,and insorbitol,and Na CI are also not sensitive in CMMedium.While,in a variety of amino acid transfer tests.when knocked out GAP will infect the absorb of animo.In summary,GAP3 gene played an essential role on vegetative growth,generation melain,pathogenicity,the absorption and utilization of hydrogen peroxide.and transport amino.
