Non-coding RNA can not encode proteins,but it can regulate living activity of organisms at the RNA level.Currently,non-coding RNAs are classified into two categories based on their length: one is small RNA with a length of 19-31 nt.The typical represent is miRNA,which complementary to transcriptional genes and result in gene silencing.The lncRNA with a length of more than 200 nt and involved in the regulation of many cellular processes such as chromatin remodeling,transcription and gene expression.They play an important role in the development of many human cancers.Recently,studies on lncRNAs in mammals is rifeness and have shown great research prospects.To explore whether nematode lncRNAs play a role in nematode innate immunity,we used RNA-seq to analyze the lncRNA expression profile of Caenorhabditis elegans infected with the pathogen Pseudomonas aeruginosa PA14 and found that compared with fed the standard food E.coli OP50,the caenorhabditis elegans infected with the pathogen Pseudomonas aeruginosa PA14 have 20 lncRNAs expression increased and 2 lncRNAs expression decreased.We speculated that up-regulated lncRNAs may regulate nematode innate immunity through some mechanism.By constructing RNA interference vectors for 17 of these up-regulated lncRNAs,and then performing RNAi experiments.We found lncRNAs TCONS00177904 and TCONS00116737 RNA interference showed increased sensitivity to Pseudomonas aeruginosa PA14,indicating that lncRNAs TCONS00177904 and TCONS00116737 are positive regulation for nematode innate immunity.The results of bioinformatics analysis showed that lncRNA TCONS00177904 can bind to miR-250 and miR-243 through base complementary pairing;lncRNA TCONS00116737 can bind to miR-45,miR-77,miR-71,miR-54,miR-73 and miR-47 through base complementary pairing.Using the above miRNA mutants to verify and found that mutations in these miRNAs did not affect the ability of C.elegans to resist P.aeruginosa.LncRNA TCONS00177904 interferes with mutant in miR-250 or miR-243,The results of resistance to PA14 are the same as lncRNA TCONS00177904 interferes for wild type Caenorhabditis elegans;lncRNA TCONS00116737 interferes with mutant in miR-45,miR-77,miR-71,miR-54-55,miR-73-74,or miR-47.The results of resistance to PA14 are the same as lncRNA TCONS00116737 interferes for wild type Caenorhabditis elegans,too.This suggests that the nematode lncRNAs TCONS00177904 and TCONS00116737 regulate nematode innate immunity do not through adsorping miRNAs to regulate mRNA.In summary,when the Caenorhabditis elegans was infected by Pseudomonas aeruginosa PA14,the expression of some lncRNAs in the nematode increased.The up-regulation of lncRNAs TCONS00177904 and TCONS00116737 enhanced the resistance of nematodes to Pseudomonas aeruginosa ? That is to say,lncRNAs TCONS00177904 and TCONS00116737 play a role in the innate immunity of nematodes.
