Proteomics,as a research hotspot,has evolved from protein qualitative analysis to quantitative analysis,and the research of proteomics has been deepening.Studies have shown that the many diseases of humans will lead to changes in the abundance of protein in vivo,so the quantitative research of protein has a very important application value.In recent years,with the increasing accuracy of mass spectrometers,proteomics has also entered a period of rapid development.Mass spectrometers with high throughput and high reliability have also become a major research field in proteomics.So how to improve the analysis efficiency of mass spectrometers has also become an important research content.Now the most widely used protein quantification method is the label-free quantitative method,which mainly includes spectral counting method and signal intensity method.Both of these methods require information on the MS/MS spectrum to quality the peptides and then quantify the protein.In this process,the spectral counting method does not use the MS spectral information.The signal intensity method uses only the information corresponding to the qualitative peptides in the MS spectrum.Therefore,a large number of peptide intensity information in the MS spectrum is not used.In view of the above problems,this paper establishes a protein quantification process based on annealing algorithm,and studies the improvement of using efficiency of the MS spectrum in mass spectrometry data.Specific innovative work is as follows:1)This paper first implements a peptide quantitative algorithm,which serves as a framework for subsequent quantitative protein research.The algorithm includes the data processing of the MS spectrum,and the peptide ion resolution is used to identify the different peptide ion peaks.The algorithm also describes how to extract XIC from a MS spectrum,while the remaining MS spectral information is used for the quantitative of peptides.2)In order to extract the information in the above-mentioned residual spectrum,we propose to extract XIC from MS spectrum using all the possible peptide ions.If a peptide can extract a valid XIC,then it is said that the unqualified XIC and peptide form a pair.In order to list all the possible peptides,this paper proposes to map the theoretical protein library into a library of peptide theory by digestion,performing a spectral search operation on the peptides in the library,and finally generating a pair of peptides and non-qualitative XICs.3)This paper presents a qualitative model of peptide matching.Through the solution of the model,the optimal pairwise pairing of the candidate pair and the unidentified XIC is obtained.In this paper,the model is incorporated into the peptide quantitative algorithm,in order to achieve an improvement of peptide quantitative algorithm.4)Peptide matching qualitative model is an optimization problem,which is solved by the use of the annealing algorithm in this paper.The effect of different optimization algorithms on matching algorithm are studied experimentally.And peptide matching qualitative model was proved that it can increase the quantitative number of peptide segments effectively.In summary,this paper follows the development of proteomics equipment and technology,focusing on analysis and mining of mass spectrometry data.This paper mainly studies the protein quantification algorithm,and develops the corresponding tools and software for the quantification of peptide and protein.The final experimental results also show that the research can achieve better results.
