Objective:To investigate the effect of different concentrations of Histone deacetylases inhibitor-Sodium butyrate(HDACi-NaB)on the expression of interferon regulatory factor 5(IRF5)gene.Methods:Human lung adenocarcinoma cell line A549 and human embryonic kidney cell HEK293 were cultured in vitro.Planting the cells,which were dealed with deionized water(group C)and NaB(group A)in the concentrations of 1.0,2.5 and 5.0mmol/L,respectively,when they were in a proper concentration.Then the promoter activity of IRF5 gene was tested by double luciferase reporter gene analysis technique.The mRNA and protein expression level of IRF5 gene were detected by RT-qPCR and Western blot.Results:Compared with group C,different concentrations of NaB in group A decreased the promter activity of IRF5 gene in A549 cells and HEK293 cells(P<0.05 or P<0.01),and inhibited the mRNA and protein expressions of IRF5 gene in A549cells(P<0.05 or P<0.01).With the increase of NaB concentration,IRF5 gene promoter activity,mRNA and protein expression were decreased.Conclusion:HDACi-NaB in vitro inhibits the expression of IRF5 gene.
