Mannose is an important functional six-carbon sugar,the main method of traditional production of mannose is hydrolysis,but hydrolysis process is unstable,causes complex separation and purification,and heterogeneous thermodynamic reaction will affect mannose formation.Therefore,the catalytic transformation of mannose-catalyzed cascade was designed here.The research of enzyme catalytic system is a popular research project,which has the advantages of mild enzymatic reaction,high efficiency and high yield.The study on the key enzyme mannitol-dehydrogenase has proved that it has higher ability to convert mannitol into mannose about 84%.Due to the involvement of Coenzyme,the two-enzyme coupling system of mannitol-dehydrogenase and NADH oxidase was constructed,which reached about 90~99% conversion rate in a short time,and solved the cost problem caused by the high coenzyme.In order to reduce costs in the process,the reaction of mannose-dehydrogenase and formic acid dehydrogenation enzyme coupling to the conversion of Mannitol was also established,and the reaction was also used to the coenzyme circulation system,which had lower cost and higher conversion rate.But in the end,the four enzymes combined catalytic reaction system was not successfully constructed.In order to find more economical and readily available raw materials,the Arabidopsis and celery-derived genes closely related to the mannitol-1-dehydrogenase gene were cloned to construct a carrot source and Arabidopsis-derived mannitol-1-dehydrogenase expression vector.
