Longan(Dimocarpus Longan Lour.)is a famous subtropical fruit tree in China.It is rich in nutrition and has high medicinal and economic value.With the development of longan industry and the change of people's preferences,the varieties of large pit,thin pulp and low edible rate have been unable to adapt to the development trend of the purpose to longan mainly in fresh,so the research on molecular mechanism of the longan seed abortive is of great significance to the breeding and production practice of longan.By comparing the data of longan transcriptome data and the related genes of arabidopsis seed,we obtained the homologous gene DIFERONIA and DICYP78A5 of longan seeds.The study is based on arabidopsis thaliana for the incomplete biological information of longan and the long growth cycle.According to the function research of arabidopsis FERONIA and CYP78A5,we obtained the arabidopsis transgenic lines by constructing the overexpression vector,rescue vector and subcellular localization vector of longan gene DlFERONIA and D1CYP78A5 to research gene function,and further explore the mechanism of longan embryo development through the differential interference contrast micrography analysis.The main results as follows:1.Overexpression DIFERONIA gene results in small seeds,small seed coat cells and small mature embryo cells,it indicates that DIFERONIA regulates seed size through cell expansion.And the siliques are smaller while there is no obvious change to the seed number of per silique;Overexpression DlCYP78A5 gene results in big seeds,small siliques and less seed number of per silique,which may be due to the reduction of fructification rate on account of DlCYP78A5 overexpression.And its branches are reduced.2.The DlFERONIA gene can rescue the feronia mutant phenotype,showing the smaller seeds,smaller seed coat cells and mature embryo cells,and smaller siliques.3.The DlFERONIA may control seed size with MINI3 and IKU2 pathways in arabidopsis;DlCYP78A5 may control seed size with IKU1 pathways in arabidopsis4.DIFERONIA and DlCYP78A5 are localized in cell membrane or cytoplasm.
