The earliest studies of the neural guidance cue netrin-1 receptor UNC5B and UNC5C were mainly focused on the development of the nervous system,and both mediated the repulsion signal of the guiding factor netrin-1 during the guidanceprocess of the Axon,which plays an important role in the development of the nervous system.Meanwhile,there are also some reports on UNC5B in the vascular system.As a receptor of netrin-1,it can repulse the vascular tipcells and regulate the vascular development process.However,the function of UNC5C in the vascular system has rarely been reported,but in the previous study work of our lab,we found that UNC5C is also expressed in the vascular system and plays an important role in thedevelopment of the vascular system.As the members of the UNC5 family,whether there are similarities in their functions in the vascular system,or whether there are complementary effects between them,so in this paper we are intends to analyze the functional characteristics of UNC5B and UNC5C in vascular endothelial cells.In order to study the role of UNC5B and UNC5C in blood vessels and lymphatic vessels,HUVEC,HUAEC and HLEC were selected as the subjects.Through in vitro tube formation experiments,we found that the expression of UNC5B and UNC5C increased significantly during tube formation process,and showed the trend of raising first and decreasing later.Then in HUVEC cells,we over expresse the UNC5B and UNC5C respectively,and conducted in vitro tube formation,migration,andproliferation experiments.The results showed that after over expressing UNC5B and UNC5C,HUVEC's tube formation ability had a considerable degree of inhibition,the same,in its ability of proliferation and migration was also inhibited,indicating that the function of UNC5B and UNC5C on the functional characteristics of HUVEC cells were similar.Further,we knocked down UNC5B and UNC5C in HUVEC,respectively,and conducted double knockdown experiments.The results showed that the cell's tube formation,proliferation,and migration capabilities were all comparable upregulated after knocking down UNC5B and UNC5C.In the double knockdown experiment,the cell's tube formation,proliferation,and migration capabilities show no significantly change compared with the single knockdown of UNC5B or UNC5C,indicating that UNC5B and UNC5C may have a functional redundancy in the control of angiogenesis.Maybe they are regulate cell tube formation,proliferation,and migration in the same way.At the same time,we conducted the same experiment in HUAEC and HLEC,and found similar results to HUVEC.Through the above experiments,we have concluded that UNC5B and UNC5C have a considerable degree of inhibitory effect on the vascular and lymphatic endothelial cell growth,proliferation and migration capabilities,and as two members of the UNC5 family,there may be functional redundancy between their functions.But the detailed molecular mechanism of its role needs to be further studied.Through the study of the functional characteristics of two molecules in vascular and lymphatic endothelial cells,may provided for the in-depth understanding of the functional association between members of the neuroguiding factor receptor family.
