Livestock and poultry germplasm resources are important for maintaining ecological security and agricultural stability.It promotes economic prosperity and social progress.The genetic resources of domestic animals should be taken seriously and protected effectively.Mesenchymal stem cells have strong self-renewal and capacity of differentiation,which provide new ideas for the preservation of livestock and poultry genetic resources.The study of mesenchymal stem cells has become a hot topic in the protection and utilization of livestock and poultry genetic resources,regenerative medicine and tissue engineering.In this study,the mesenchymal stem cells derived from the broiler skin and goose heart were taken as experimental subjects,and the cells were isolated,cultured and induced into other types of cells in vitro.The following results are obtained.1.The broiler DMSCs was isolated with collagenase I and trypsin and cultured for 31 generations in vitro.The cell morphology was mainly long spindle shape.The results of RT-PCR identification showed that the cells expressed CD73,CD29,CD44,and CD71.The results of immunofluorescence showed that CD105,CD90,CD73 and CD44 were positive expression in broiler DMSCs.The result of flow cytometry showed that CD29,CD44 and CD73 was highly expressed in broiler DMSCs.The growth curve of DMSCs showed a typical "S" shape,which in accordance with the general rule of cell proliferation in vitro.The population doubling time and the ability of clone-forming showed that the cell proliferation ability was decreased with the increase of generation time.Broiler DMSCs was successfully induced into osteoblasts,chondrocytes and adipocytes under certain conditions.Those can be detected by specific staining and RT-PCR identification.2.The goose HMSCs was isolated by collagenase II and trypsin,and was cultured in vitro for 21 generations,and the morphological features of the cells were spindle shaped.The results of RT-PCR and immunohistochemical detection all showed that the cells expressed specific MSCs genes such as CD29,CD34,CD73,CD166 and CD44.Through flow cytometry analysis,CD29,CD34,CD73,CD166 and CD44 were highly expressed in goose HMSCs.The growth curve of HMSCs showed that cells experienced a short latency period,entered the logarithmic phase of rapid growth,and then entered the stable phase and decline phase,showing a typical "S" shape,which in accordance with the basic rule of cell proliferation in vitro.The results of population doubling time showed that the cell proliferation ability was gradually decreased with the increase of generation time.The HMSCs were induced by adding appropriate inducer in the medium and the results were detected by specific staining and RT-PCR.The results showed that HMSCs were successfully differentiated into osteoblasts,adipocytes and chondrocytes,indicating that the goose HMSCs have differentiate ability and can be induced into multiple directions.In summary,broiler DMSCs and goose HMSCs were successfully isolated and cultured in thisstudy,and the biological characteristics and capacity of differentiation were studied.The results showed that both broiler DMSCs and goose HMSCs showed typical biological characteristics of mesenchymal stem cells.The experimental results are of great significance for the preservation of livestock and poultry germplasm resources,and provide experimental basis for regenerative medicine and tissue engineering.
