SOX9 is an important member of the SOX(SRY-related HMG box)family.The human SOX9 gene is located on chromosome 17q24.3-q25.1 and encodes 509 amino acids.It is a transcription factor involved in gonadal development.Recent studies have showed diversity role of SOX9 gene.As a transcription factor,SOX9 participates in the development of embryos,and plays a key role of sex determination and the formation of cartilage.However,so far,many biological functions and mechanisms of SOX9 remain to be elucidated.WD repeat-containing protein 5(WDR5)is a member of the WD-40 protein family,which is a 40-amino acid conserved sequence.WDR5 is an essential component of the MLL(mixed lineage leukemia)complex,which triggers histone methylation at lysine 4 of histone3 preferentially as an active epigenetic mark at 5' coding region of the gene.Recently,it was found that WDR5 and SRY can co-localize on the SOX9 promoter and SRY cooperated with WDR5 in inducing the expression of SOX9,which could be important for sex determination.However,there is still no known how SOX9 interacts with WDR5.Therefore,using the gene cloning the over-expression vector of SOX9 gene were constructed.Successfully established the LNcap cell strain stable expression of SOX9 aiming for the effect of SOX9 on WDR5 by Chromosome immunoprecipitation(Ch IP).
