Abstract/Summary:
Microbial resources are one of the important sources of drug development at this stage.Plant endophytic fungi have unique metabolic mechanisms because of their unique living environment,and their metabolites show a variety of chemical structures and biological activities.Research on secondary metabolites of plant endophytic fungi plays an important role in the development and utilization of microbial resources and the search for pharmacologically active compounds.The species used in this study was Aspergillus oryzae,which is an Aspergillus genus and its distribution range is very wide.It is also an important industrial fungus.Current research shows that the secondary metabolites of Aspergillus oryzae contain various chemical constituents such as alkaloids,terpenoids,flavonoids,steroids,and coumarins,and have a wide range of pharmacological activities,including anti-tumor and anti-tumor activities.HIV,anti-oxidation,antibacterial and antibacterial activities.In this study,OSMAC(one strain-many compounds)methods were used to screen the optimal culture conditions for Aspergillus oryzae strains.First,change the culture conditions(different culture medium),add precursors(D-tryptophan),add enzyme inhibitors,etc.,were used for small sample fermentation,followed by thin layer chromatography and HPLC analysis,the final selection of liquid amplification fermentation(spot culture medium and D-tryptophan)and solid amplification fermentation(Rice,perlite and water and vorinostat deacetylase inhibitor Agents)two methods to ferment Aspergillus oryzae strains.28 monomer compounds were isolated from their fermentation products,including 4 new alkaloids and 24 known compounds.they are,respectively: Aspergillin N(1*),Aspergillin O(2*),Aspergillin P(3*),Oryzae A(4*),Indole-3-methylethanoate(5),12-N-methylcyclo-(L-tryptophyl-L-phenylalanyl)(6),Ditryptophenaline(7),1H-indole-3-carboxaldehyde(8),Indole-3-carboxylic acid(9),Indole-3-acetic acid(10),N2′-Acetyltryptophan methyl ester(11),1-(2-oxopropyl)indole-2,3-dione(12),Luteolin I(13),2,3-dihydro-4(1H)-quinolone(14),4-hydroxybenzoic acid(15),p-hydroxypheny lacetic acid(16),Methyl 4-hydroxyphenylacetate(17),Speradine B(18),Aspergilline D(19),Speradine C(20),Cyclopiamide D(21),Speradine D(22),Cyclopiamide B(23),Cyclopiamide F(24),Speradine G(25),Cyclopiamide E(26),Phomaligol A(27),1-(3,4-dihydroxy-5-methoxyphenyl)propan-1-one(28).
