MLKL monomer formed octamer is an essential step in necroptosis.Also,octamer is an important unit for cell to execute necroptosis.However,the mechanism of forming MLKL octamer is still largely unknow.Here we show that the MLKL oligomer is mainly octamer.And individual MLKL could form octamer via core amino sites.MLKL octamer is composed by tetramer.Besides,MLKL could not form a larger oligomer rather than octamer.In the in vitro experiment,I found that MLKL can only form a single peak of oligomer.during the formation of MLKL octamer from tetramer,some key animo sites are very important.When we mutated the S79 to A,D106 to A,MLKL can not form octamer from tetramer.When we mutated T122 to c,we could see an octamer band in western withour DSS.These data show that S79,D106,T122 have significant role in necroptosis.We also found that R30,R34 have crucial function when MLKL binding to membrane.In addition,when MLKL binding to cell membrane,the C-terminal and N-terminal is still in cell plasma.Our data illuminated the detailed of MLKL function in necroptosis.
