Transmembrane protein(TP)is a type of protein that is embedded in the biological membrane.Transmembrane proteins play various important function in cell though they usually have a low level of expression.A bunch of extracellular signaling receptors like EGFR and TGF?R are transmembrane proteins,while other transmembrane proteins play a role in moving a specific substance through the membrane.Ms9 is a sigle bitopic transmembrane protein.Studies about Ms9 are rarely reported,the exactly structure,location,fuction of Ms9 still remain unknown.Our study devote to explore the location and modification of Ms9,whether Ms9 could be secreted and the influence on EGFR signaling pathways by overexpressing Ms9 with tags.We found that Ms9 is located on intracelluar membrane and plasma membrane,Ms9 is a N-lined glycoprotein and it could be secreted to extracellular environment for its biological function.Ms9 could interact with EGFR as well as other receptors like TGF?R and TLR4,much more than this,Ms9 could suppress abnormal EGFR signaling activation due to extracellular stimulation to a certain extent.Ms9 could also bind to itself although the mechanism remains unclear.Our study shows that Ms9 could suppress unusal EGFR signaling up-regulation in normal cells due to the secretion of cancer cells.Thus,it is possible that new drugs on the basis of Ms9 could suppress tumor development in cooperation with other therapy ways.
