The Bacillus spp.is reprised of gram-positive,spore-forming,were used in agriculture,industry,medicine,environment because of their strong adapting ability with stress-resistance spore.Qinghai Tibet Plateau is the highest plateau in the world,with an average altitude of over 4000 meters.Due to its unique extreme environment such as low temperature,hypoxia and strong radiation,etc,special Bacillus species may be bred in this region.In this study,8 soil samples from the Qinghai Tibet Plateau were obtained from the Zhu Haiyan Research Institute of the Chinese Academy of Sciences,respectively,and 8 Antarctic soil samples were obtained from the Zhu Renbin Laboratory of University of Science&Technology China.There were 84 strains of Bacillus were isolated from the 16 soil samples,84 isolates were identified with biochemical method and molecular identification including ERIC-PCR fingerprint analysis,16S rDNA and gyrB gene sequence analysis.The results showed that 25 Bacillus strains were identified into 7 isolates of Bacillus amyloliquefaciens,9 isolates of B.simplex,1 isolates of B.pumilus,5 isolates of B.licheniformis,1 isolates of B.thuringiensis,1 isolate of Lysinibacillus sp.and a new strain of Bacillus which needs further research.Isolation and identification of the strains for the prevention and control of maize against the top rot of Bacillus screed provides a source of bacteria.Corn production and consumption is large in China.In recent years,the occurrence of bacterial diseases in maize is serious in some areas,such as Hebei and Shandong Province.This event has brought serious harm to agricultural production and food safety.We collected maize top rot plants isolated four strains of pathogenic bacteria from Shandong three areas(Jiyang,Taian Ma Zhuang,Zibo),according to 16S rDNA alignment results identified both as Pantoea ananatis.And according to the comparison of gyrB gene results are identified as P.ananatis,namely maize top rot pathogen for pineapple P.ananatis.This study isolation and identification of 25 strains of Bacillus by flat antibacterial experiment from the Qinghai Tibet Plateau and the Antarctic soil screened 4 strains of antibacterial effect of good strains of Bacillus,respectively EZ15-08,EZ01-05,EZ15-07,EZ15-09.Among them,the best antibacterial effect is the strain EZ15-08.
