The Symbiotic nitrogen fixation between rhizobia and legume plants is an important way to get nitrogen in the nature.The researchers had cloned a series of key symbiosis genes by using the different symbiotic defect mutants of model legume(Medicago truncatula and Lotus japoncius).Based on these results,researchers established the model of nodulation signaling transduction pathway.In order to establish the rice-Rhizobium symbiotic nitrogen fixing system,we transfer these key symbiotic genes into rice and obtained transgenic rice.After we found the expression of these genes can affect the rice related signal pathway by rhizobium inoculation,we used rice cDNA microarray to detect the root transcriptional group of transgenic and control group.Both of the experiments were under the condition of 7 days after inoculated and uninoculated.The main results are as follows:1.After the T0 generation transgenic rice was obtained,we get a large number of single copy homozygous transgenic rice by identifying and screening.2.Repeat verifying the early results of phenotypic observation: Ten days after inoculation,both transgenetic group and control can observe rhizobium invasion of the epidermis and root hairs;rhizobium inoculationincreased the number of lateral root and the lateral roots number of transgenic group was significantly greater than the control group.3.Expression of genes were changed related to plant hormone signal transduction pathway.Comparison of transgenic group and the control group,the genes related to auxin signaling pathway and cytokinin signaling pathwaywere up-regulated4.Expression of genesrearrangementrelated to plant defense response.Both transgenic and inoculation can significantly increase the expression of genes related to plant defense response.
