Microbial resources are very abundant in the ocean.Marine microorganism have different metabolic pathways under the influence of diversity and particularity of the environment,which can secrete a variety of novel and active substances.Most of the active substances in the chemical,pharmaceutical,food and life science and other fields have a broad application prospect.Aspergillus sp.AA 005 were isolated from the samples collected from hydrothermal vents in Taiwan Kueishantao.They were cultured in two ways,which in both common condition and under metal stress(Cu-400 ?mol/L,Ni-100 ?mol/L,Mn-400?mol/L).We are to explore the synthesis of the secondary metabolites of Aspergillus sp.AA 005.The chemical compositions of the extractive were separation and purification by using TLC,Positive phase silica gel chromatography column,HPLC,SephadexLH-20 gel chromatography,polyamide column chromatography and other modern chromatography technology.The relative molecular mass of compounds were analyzed by MS,and the structures were identified by NMR spectral data and other methods.Fourteen compounds in total were got from the strain Aspergillus sp.AA 005,among which two compounds were prove novel.Secondary metabolites were obtained from Aspergillus sp.AA 005 in common condition:Ergosta-7,22-dien-3?,5?,6?-triol(W-1),(22E,24R)-ergosta-5,7,22-trien-3?-ol(W-2),Sterigmatocystin(W-3),3,3'-dihydroxy-5,5'-dimethyldip henyl ether(W-4),Cerebroside A(W-5),Kojic acid(W-6),3-O-methylviridication(W-7),Succinic acid(W-8),Thymine(W-9).Secondary metabolites were obtained from Aspergillus sp.AA 005 under Cu-400 stress:8-Hydroxy-3-O-methylviridation(W-10),6-Methoxy-3-O-methyl viridation(W-11).Secondary metabolites were obtained from Aspergillus sp.AA 005 under Ni-100 stress:Cordyol C(W-12).Secondary metabolites were obtained from Aspergllus sp.AA 005 under Mn-400 stress:Macrolactone(W-13),3-(3-methoxy-5-methylphenoxy)-5-methylphenol(W-14).There are two new compounds,8-Hydroxy-3-O-methylviridation(W-10),6-Methoxy-3-O-methylviridation(W-11).Their structures of two stressed products showsed no similarities with common metabolites of the strain.Aspergllus sp.AA 005 under metal stress will synthesize some new secondary metabolites.It is proved that the stress culture of metal ions training to fill the content of OSMAC theory,and is of great significance for the search of new marine secondary metabolites.
