Polyacrylamide is widely used in oil extraction process,and the amount is increasing day by day.Recently,studies have found that when the polyacrylamide is slow degradationed,it may product acrylamide(AM),and the other oligomer in the environment.AM were widely used in industrial production,includ the waste water treatment,paper making,textile.People found that the food after high temperature processing can produce large amount of AM.Studies have shown that AM is a moderate toxic material,it has the nerve toxicity,genetic toxicity,reproduction toxicity and carcinogenicity,etc.Due to there is no clear standard to contror AM run into the environment,it may have potential toxicity to human.Study about AM toxicity for people is very urgent and necessary.Zebrafish as the model organisms in toxicology research is very rational.Zebrafish is translucent,vitro fertilization and development,can observe the change of each stage developmenlt directly.After fertilization 24h,the main organs primordium has formed,each ventricle,eyes,ears and blood cells,segment can clearly observed,development rapidly,rely on absorb yolk sac can survive until hatch,can observed the teratogenic effect caused by abnormal forms directly,can observed the testing end in toxicology tests.Using the gradient concentration method in this experiment,observe and counting the deaths zebrafish embryo under the effect of AM,calculate the 24h,48h,96h median lethal concentration of zebrafish embryo live with AM,96h median lethal concentration as a parameter,for a long-term toxicity research,observe the zebrafish embryo exposed in different concentrations of AM,observe its voluntary movement frequencyand the heart rate,clear the effect of zebrafish embryo exposed AM at the beginning and organ formation stage,through the investigat for the hatching rate and deformity rate,clear the influence of overall development of the zebrafish embryo exposed in AM;through the morphological observe,can directly reflect the AM toxic effect of zebrafish embryo,observed the effect of zebrafish embryo different stage of development exposed in different concentrations of AM.Under normal circumstances,the organism can produce a lot of free radicals,small amount of free radicals is indispensable to the body,the excessive free radicals can cause oxidative damage to the body,affect the normal function of the body,SOD can remove excess free radicals,keeping the balance of the organism,when the organism caused by oxidative damage,MDA content increased,through test the activity of SOD and the content of MDA in zebrafish embryo,clear the degree of zebrafish embryo from oxidative damage caused by AM.The experimental result shows that the 24h median lethal concentration of AM of zebrafish embryos is 377.04 mg/L;the 24h median lethal concentration of AM of zebrafish embryos is 214.6 mg/L;the 24h median lethal concentration of AM of zebrafish embryos is 192.58 mg/L.zebrafish embryo exposed in AM for 24h,the frequency of autonomous motion is increased,zebrafish embryo exposed in AM for 48h,the frequency of heart rate is increased,the AM have toxic effect on zebrafish embryo early embryonic development,zebrafish embryo exposed in AM for 72h,the hatchability is reduced,AM exposure has hatched zebrafish appear deformity,the reasult shows that AM have toxic effect on the overall development of zebrafish embryo;through the morphological observation,exposed in AM can formed such as cardiac oedema,scoliosis,stop developmental and malformation.The reasult shows that the SOD activity of zebrafish embryo is decreased,the content of MDA is increased,the zebrafish embryo received oxidative damage.Through the experiment reserch,the result shows that the AM effect the organ formation stage,by reduce the zebrafish embryo hatching rate,increase the frequncy of deformity,influence the overall development of zebrafish embryo;the SOD activity of zebrafish embryo is decreased,the content of MDA is increased,prove that the zebrafish embryo received oxidative damage.
