Since avian influenza virus was found,avian flu emergs consistently around the world.The first case of human infection of H5N1 avian influenza virus happened in 1997.All this has brought a great threat to humans and the poultry industry.In recent years,the avian influenza virus is mutating,there has been some new features.Such as the waterfowl infection generally not develop the disease which is bccause they are the natural reservoir,but now the new avian flu virus can cause morbidity and mortality of waterfowl.In addition,incidence birds will show symptoms of neurological disorders,and we can isolate the virus in the brain,which in the past not hanppened.Zou inoculated chickens with neurovirulent avian influenza virus,finding typical virus non-suppurative encephalitis in brain,with a large number of astrocytes dying.Astrocytes provide the necessary nutrition to neurons in the nervous system,maintain ion balance around neurons and involve in the regulation of neuronal activity,which are importment part of the nervous system.Besides,disfuction of astrocytes is closely related to stroke,Alzheimer's,epilepsy,and spinal cord injuries etc.,and play an important function in the process of synaptic generation and damage repair.To explore neural pathogenisis of avian influenza virus,the study went on from two aspects:1.Genomic expression profiling in neural cells infected with neurovirulent avian influenza virusUsing genomic expression profiling in neural cells infected with neurovirulent avian influenza virus,we can find lots of DE genes.Through classification and function analysis of DE genes,we discover that DE genes particitate in neural signal transduction regulation,innate immune response,inflammation and acute phase response.It should be noted that some DE genes involve in axon morphology and function stability,and some in the pathogenesis of Huntington disease process.2.Nerve invasion pathway of avian influenza virusNeurovirulent avian influenza virus is able to enter central nervous system through the outer peripheral nerve.But the path of destruction of the blood-brain barrier through the blood circulation has not been confirmed.In this experiment,using the characteristic of Evans blue binding albumin,we can study the integrity of the blood-brain barrier of mice with neurological symptoms.The results show that though neurological symptoms of mice is particularly obvious,the integrity of the blood-brain barrier is still unspoiled,which show that avian influenza virus can not enter the CNS through the blood circulation path,providing some reference for the neurological pathogenesis of avian influenza virus.
