AXIN is multifunctional scaffold protein is involved in many signal pathways,such as JNK signal pathway,Wnt signal pathway and mTORC signal pathway.Recently,we took the yeast two-hybrid system as a tool for searching candidate proteins that could interact with AXIN,and isolated a clone encoding LAMTOR1(late ensosomal/lysosomal adaptor,MAPK and mTOR activatorl).Ragulator,consisting of LAMTOR1 to 5 is an essential pentamer complex for mTORC1 activation.Based on the previous studies that show AXIN is an important factor which can facilitate AMPK(AMP-activated protein kinase)sensing energy levels,we speculated whether LAMTOR1 also be involved in this pathway.For futher research,we established the liver LAMTOR1 knockout mice.We found that the liver LAMTOR1 knockout mice was abnormal not only in fat metabolism and AMPK activation levels after fasting,which was similar to the liver AXIN knockdown mice,but also in gluconeo genes is.My work has thus provided a fundamental support to the research of the functions of LAMTOR1,AMPK and Axin.
