Activin is a member of transforming growth factor-beta(TGF-?)superfamily,and a multifunctional growth and differentiation factor.The role of activin has distinct tissue specificity.Because the Smads family proteins can't explain this phenomenon in the activin's signal transduction pathway,we research the functions of ARIPs(Activin Receptor Interacting Proteins).As the ARIPs members,ARIP1-5 can interact with activin receptor II at their N regions,thus the key point to understanding the differences among these different activin signal pathway is explaining the functions of ARIPs'C region.Yeast two-hybrid system is an effective experiment technique to find the interaction between protein and protein.In order to further investigate the functions of ARIP5 in the activin signal transduction pathway,we used pGBK-ARIP5 as the bait protein and screened the human fetal cDNA library through the yeast two-hybrid system.Fifty yeast positive clones were obtained,in which extracted plasmids,and then used these plasmids transformed the E.coli.We sequenced the insert segments from the positive clones which can grows on the LB/Amp plate,then we analyzed the sequence result.We used the technique of yeast two-hybrid system found the proteins which can interacted with ARIP5 may completed the network of activin's signal pathway.Our experiments may contribute to the further understanding of the activin's function and provided new therapy for the diseases which relates with activin.
