Soil salinization is a major limiting crop productivity conditions,because it not only reduces the production of crops,but also limits the development of agriculture on uncultivated land.Plants can take advantage of Na+/H+ antiporter mediated Na+ efflux and Na+ segment maintain low Na+ concentrations of the cytosol.Located on the plasma membrane Na+/H+ antiporter protein SOS1(salt overly sensitive 1)can excrete excessive Na+ out of cells,reduce the toxic effects of Na+.Bruguiera gymnorrhiza is one kind of mangroves,it has a strong salt tolerance,so Bruguiera gymnorrhiza should be a kind of excellent material to study the salt tolerance.On the basis of predecessors,the mutant gene BgSOS1-DAPA and BgSOS1-3000 were transformed into yeast mutants AXT3K,and further explore the regulation mechanism of protein BgSOS1.The main results are as follows:1.To amplify the sequence of Seri 143 and Ser1145 mutations of BgSOS1(BgSOS1-DAPA)by PCR,constituting yeast expression vector was transformed into the salinity sensitive strain AXT3K.To amplify the sequence of the deletion mutant BgSOSI-3000 by PCR,constituting yeast expression vector was transformed into the salinity sensitive strain AXT3K.The results show that either single AtSOS2 or the SOS2-SOS3 protein kinase complex both could regulate the activity of BgSOS1?BgSOS1-DAPA.We also found that BgSOS1-3000 grow well at a high salt concentration,this means that BgSOS 1-3000 has a high tolerance of salt.2.We research the situation of CBLs interact with SOS2 to regulate BgSOS1,The results show that CBL 5,CBL 8,CBL 9 and CBL10 can interact with SOS2 to regulate BgSOS1.We selected CBL10 interact with SOS2 to regulate BgSOS1-DAPA and BgSOS 1-3000,the results show that CBL10+SOS2 can regulate BgSOS1-DAPA.3.We selected 12 members of CIPKs to regulate BgSOS1,constituting yeast expression vector was transformed into the salinity sensitive strain AXT3K.We found that only CIPK24(SOS2)Can regulate BgSOS1.
