Nucleo-cytoplasmic transport is essential for eukaryocyte to maintain its life activity.Importin-P type nucleus transport receptors are able to mediate variety of cargos with specific functions to import into nucleus,and this kind of regulation substantially exist throughout a life process.In this thesis,we cloned Solanum lycopersicum transportinl?SlTRN1?.Amino acid sequence comparisons show that SlTRN1 is one of importin-Ps with relativly conservative sequence.The similarities between SlTRN1 and the TRNIs of Arabidopsis thalina,Populus trichocarpa,Citrus sinensis,Citrus clementina,Theobroma cacao,Prunus mume or Ricinus communis are all more than 70%.The expression of SlITRN1 in Arabidopsis thalina can recover the phenotype of the sicl-1 mutant,such as increasing the stem length,reducing the number of branches,elongating the internode and recovering the leaf area.In root cells of Arabidopsis thalina,SlTRN1 is mainly localised in nucleus and also can be observed in cytoplasm.Yeast two-hybrid results show that SlTRN1 can interact with some of those cargos of AtTRN1 including AtlG56280,AtlG61170,At2G24790,At2G39730,At3G44310,At3G50330 and At4G39140,however,there is no interaction with At5G24930,At5G 14040 or At4G38710.This result indicates that the recovery of sicl-1 mutant is caused by the restoring of nucleo-cytoplasmic transport by SlTRN1.In Solanum lycopersicum,we cloned eight homologous proteins of those cargos of AtTRN1 in Arabidopsis thalina.Yeast two-hybrid test shows that five of these homologous proteins,LOC101262262,LOC101255184,LOC101252594,NM001247087 and LOC101055547,can interact with SlTRN1,and all of them contain M9,NLS or PY-NLS sequences.These results indicate that the reconition between SlTRN1 and its cargos is conservative.SlTRN1 and AtTRN1 can import similar cargos into nucleus.Different species of plant may take the same strategy to recognise and import cargos using TRN1,and TRN1 may maintain its functional conservation during evolution.
