Object: Liver is an important metabolic and immune organ in the body.The incidence of liver injury is on the rise,and the mortality is high.It has become a major threat to human health.Research shows that activin is involved in the induction of liver injury,activin is transforming growth factor beta(transforming growth factor,TGF-3)new members of the superfamily,investigated the effects of activin A(Activin A)and activin interacting protein 5(activin receptor-interacting 5 protein,ActRIP5)in concanavalin A(concanavalin A ConA)expression and significance of acute immune liver induced injury.The animal,which lays a foundation for clinical prevention of hepatic fibrosis gene seeking new targets.Methods: C57BL/6 mouse tail vein injection of ConA(15mg/Kg),the change in different periods respectively after administration in batches of serum enzymes of change,animal model of liver and spleen index and liver tissue pathology and morphology,the expression of Activin A and ActRIP 5 was detected by fluorescence quantitative PCR.Results: The serum levels of ALT,AST of liver injury model began to increase 4h after ConA injection and reached the peak in 24 h,then gradually decreased,but has not yet returned to normal level in 96 hours.The spleen index began to increase 2h after administration and reached the peak after 24 h,and began to decline after 48 h,and returned to normal level after 96 h.The liver index began to increase 24 h after administration and reached the maximum after 48 h,then began to decrease,and didn't yet returned to normal after 96 h.The test of morphology and pathology revealed liver injury began to appear 4h after administration and was most serious during 24h-48 h,and began to recover after 48 h,but did not yet return to normal level after 96h;Fluorescence quantitative PCR results showed that Activin A began to decline 2h after administration and reached the peak after 8h,then began to decrease,and decreased to normal level afeter 96h;ActRIP5 began to rise 4h after administration and reached the peak after 8h,then began to decline,but did not return normal levels after 96 h.Conclusions: ConA can induce acute liver injury,and the expression of Activin A and ActRIP 5 abnormally elevated during the progression of liver injury and decreased and returned to normal during the recovery period of liver injury,suggesting that Activin A may participate in acute liver injury induced by ConA through the specific signal transduction protein ActPIR5.
