In the past ten years,bariatric surgery has become one of the standard methods for the treatment of obesity and Type 2 diabetes mellitus.The effect of sleeve gastric on weight reduction and metabolic complications compared with other weight loss surgery is best.In this experiment,ZDF rats were used to establish a model of sleeve gastrectomy.The basic physiological indexes of rats were measured at 2 weeks before operation,2,4,and 8 weeks after operation,including weight,food intake,fasting blood glucose and oral glucose tolerance.Ninth weeks to obtain rat liver,kidney,testis and pancreas tissue,formalin fixed,paraffin embedded tissue pathological changes were observed by HE staining,to observe the degree of fibrosis by Masson staining,S100 protein immunohistochemistry rats by changing the biological process of calcium signal transduction pathway.It was found that sleeve gastric volume reduction effectively relieved the body weight,blood sugar and blood glucose regulation ability of diabetic obese rats,and the spermatogenesis ability of rats was increased,the number of islet cells increased,and the renal injury was restored to some extent.Sleeve Gastrectomy(SG),as the most effective bariatric surgery was used to chronically lose weight and control glucose metabolism in Type 2 diabetes mellitus patients.In this study,visceral lipid metabolism and energy metabolism were investigated in Zucker diabetes fatty(ZDF)rats after SG.whole-body metabolic parameters were obtained through TSE LabMaster.Blood lipid and renal function were analysized by serum from rats'tail veins.The expression of renal genes was detected by real-time PCR,and western blotting was employed to detect the level of AKT/PI3K in kidney.Compared to controls,body weight of ZDF rats significantly reduced,and glucose homeostasis improved as well as energy metabolism including RER,energy expenditure at night and activity of animal.Meanwhile,serum lipid decreased,and renal function recovered.Histology analysis confirmed that ZDF rats after SG obviously decreased the size of perirenal adipose,effectively stimulated up-regulation of lipogenesis genes,while down-regulate expression of adipogenesis genes in kidney.In addition,p-PI3K and p-AKT in kidney were significantly decreased in SG group.Conclusions:ZDF rats after SG surgery improved weight loss,food intake,fasting plasma glucose and glucose tolerance of ZDF rats,which was coincident with the results of energy metabolism.SG decreased the renal fat volume,which was associated with activation of PI3K/Akt signaling pathway.This may help us better understand the mechanism of bariatric surgery contribute to kidney.
