The metal iridium complexes was widely used,such as catalysis,photochemical sensing,light-emitting diodes(LEDs),fluorescent probe and the imaging in vivo due to their excellent photo-physical properties(long fluorescent lifetime,high quantum efficiency,absorption spectrum and fluorescence spectrum is adjustable,large stokes shift and photostability).The absorption of traditional iridium complex was mainly located in the visible wavelengths between 400 and 600 nm,which has the small penetration depth.The application of near infared molecules in vivo imaging has been received more and more attention.In this paper,iridium complexes with cyanine based ligand were designed and synthesized,which presented strong absorbance at about 800 nm.Furthermore,the modified water-soluble nanoparticles of iridium complexes with good biocompatibility were well obtained.The application of nanoparticles in photoacoustic imaging of lymphatic metastases was further explored.The specific research contents are as follows: 1.Synthesis and Properties of Near-Infrared Metal cyanine-Iridium Complexesand its Water-Soluble Nanoparticles.We considered the photodynamic effects of iridium complexes and the near infrared absorption property of cyanine dye to design and synthesize the iridium complexes.The hydrophilic iridium complexes nanoparticles were obtained by coating with bovine serum albumin(BSA),which was then conjugated with glutaraldehyde in order to enhance the stability of nanoparticles.The sizes of obtained nanoparticles are around 100 nm.In vitro phosphorescence imaging and endocytosis properties of nanoparticles were studied using confocal scanning laser microscopy and flow cytometer.2.Application of Water Soluble Near Infrared Metal Cyanine-Iridium ComplexNanoparticles in Lymphatic Metastatic Photoacoustic ImagingSentinel lymph nodes are the first lymph nodes that metastasis during tumor growth,indicating the state of regional lymph nodes.Recently,sentinel lymph node biopsy(SLNB)is a normal method to determine whether the axillary lymph nodes in breast cancer patients have cancer metastasis.Until now,the lymphatic tracer meilan and nuclide are used in the operation and cannot be used for preoperative detailed evaluation.Therefore,a safe and convenient preoperative sentinel lymph node tracer method is urgently needed in clinic.The synthesized water soluble metal cyaine-iridium nanoparticles have good biocompatibility can be considered as a tracer in guiding lymph cleaning experiment.In this paper,we selected three kind of cells(4T1 breast cancer cells in mice,MDA-MB-231 human breast cancer cells and HeLa human cervical cancer)through foot pad subcutaneous inoculation to establish the lymphatic metastasis model of Bablc mouse,respectively.Diagnosing lymph node metastasis by palpation,PET-CT examination and HE staining technique were used to confirm the lymphatic metastasis of tumor.The detailed studies have shown that the obtained lymphatic metastasis was well detected by using in vivo photoacoustic imaging of nanoparticles,which is helpful for evaluation of lymph node status before surgery.It is useful for the cleaning operation of lymph node that the synthesized nanoparticles could be recognized by naked eyes.The evaluation of the status of lymph nodes before and after operation with the guiding of photoacoustic imaging is significance to clinic.
