Alzheimer's disease(AD)is a degenerative disease of the central nervous system.It has the characteristics of the onset of Alzheimer's diseases and the development process is gradual.No effective treatment has yet been found.With the aging of the population,the huge public health cost brought by AD will be an issue that each country urgently considers.The pathogenesis of AD is complex,and the A? cascade theory is currently recognized by most people.In recent years,studies have shown that cholesterol metabolism,autophagy dysfunction and the incidence of AD have a great correlation,our group in the early electro-acupuncture intervention to effectively reduce A? levels,and the use of transmission electron microscopy found that the model mice increased autophagic bubble and Most contain lipid droplets.As an important apolipoprotein in the brain,apolipoprotein E(ApoE)is involved in the metabolism and distribution of cholesterol in the brain.The relationship between ApoE and the onset of AD has become the focus of research.In this experiment,the level of A? 1-42 in hippocampal cells,the levels of ApoE4 and ApoE2 in hippocampal cells,and the level of cholesterol-rate-limiting enzyme,cholesterol-24S-hydroxylase(CYP46),were determined by enzyme-linked immunosorbent assay.Observing the metabolism of cholesterol in the brain,the decrease of A? levels may be related to the improvement of cholesterol metabolism in the brain.To explore whether the mechanism of prevention and treatment of AD by acupuncture is to improve the level of ApoE in the brain,improve the disorder of cholesterol metabolism in the brain,and then.improve the disorder of autophagy,thus reducing the level of AP in the brain.ObjectiveObserve the effect of electroacupuncture on amyloid protein(AP)and apolipoprotein E(ApoE)levels in hippocampus of 4-month-old APP/PS1 double transgenic mice,and to explore whether the mechanism of acupuncture alleviating AD is Within the level of ApoE,reducing the level of brain A?1-42.MethodFourteen male 4-month-old APP/PS1 double transgenic mice were randomly divided into model group and electro acupuncture treatment 7 for each group.C57BL/7mice with the same age were used as normal control group(n=7)."Yongquan" or"Baihui"the same conditions as a means of intervention,each lasting 15min.every other day,a course of 2 weeks.The levels of A?1-42,ApoE2,ApoE4 and cholesterol-24S-hydroxylase(CYP46)in hippocampus were measured by enzyme-linked immunosorbent assay(ELASA).Result1.The content of hippocampal A?1-42 in the model group was 74.62±12.01(pg/mg)higher than that of the normal control group 41.12±2.76(pg/mg)(P<0.01);the content of A?1-42 in the acupuncture group was 11.93±5.02(pg/mg)was lower than the model group(P<0.01).It is suggested that electroacupuncture can effectively reduce the level of A?1-42 in hippocampus of APP/PS1 double transgenic mice.2.The level of hippocampal ApoE2 in model group was 136.26±28.56(?g/g)compared with 144.37±12.75(?g/g)in normal control group,the difference was not statistically significant.The level of hippocampal ApoE4 in model group was 94.96±38.19(?g/g)was lower than the normal control group 116.66±33.06(?g/g),(P<0.01);the level of hippocampal ApoE2 in the acupuncture group was 208.16±31.86(?g/g),and the level of ApoE4 was 161.26±19.01(?g/g)are higher than the model group(P<0.05).It is suggested that the level of ApoE4 in the mouse model after the gene double conversion is reduced,electroacupuncture can increase the level of ApoE2,ApoE4 in the hippocampus,and affect the cholesterol metabolism in the brain.3.The level of CYP46 in the normal control group was 623 63±107.06(pg/mg)higher than that of the model group 567.65±130.79(pg/mg),and the CYP46 level of the acupuncture group was 583.11±145.51(pg/mg)higher than that of the model group 567.65 ±130.79(pg/mg),but the difference was not statistically significant,suggesting that the cholesterol metabolism level of the model mice after the double-turn of the gene was reduced,and the level of cholesterol metabolism in the brain could be increased after acupuncture.ConclusionElectroacupuncture can reduce the level of A?1-42 in the hippocampus of APP/PS1 double transgenic mice at 4 months of age.The mechanism may be achieved by affecting the level of ApoE in the brain.Electroacupuncture may affect the cholesterol metabolism in the brain.Process,but further research is needed.
