Experimental And Clinical Study On Yishen Jianpi Recipe In The Treatment Of Male Oligospermia

Part one:The experimental study on the treatment of oligospermia by Yishen Jianpi RecipeOBJECTIVE:In this study,the oligoasthenozoospermia mouse model prepared with reproductive toxin tripteiygium wilfordii polyglycosides was used as the observation subject,with the treatment group given Yishen Jianpi formula and the control group given(Dongweili or Zuokating group)a more generally accepted treatment medication namely L-Carnitine oral solution as treatment after model preparation.Using the resulting indicators gathered from the routine semen test,mitochondrial transmembrane potential and mitochondrial pemeability transition pore on mouse model subjected to treatment,the efficacy of Yishen Jianpi Forumula on abovementioned oligoasthenozoospcrmia mouse model was observed from different levels and standpoints.Modern scientific research concepts were utilized to prove the efficacy of Yishen Jianpi Formula for the treatment of male oligoasthenozoospermia via backing with experimental data so as to provide factual basis for the TCM method-"Yishen Jianpi" in the treatment of male oligoasthenozoospennia;as well as to provide new ideas,methods and foundation for further research in the treatment of male oligoasthenozoospennia.METHOD:50 white male BALB/c mice first fed for 5 days then randomly separated into 5 groups,namely the normal group,model group,control group,low dosage TCM group and high dosage TCM group.Every mouse weighed between 18-20g.Preparation of model mice:8ml/kg/d of Tripteiygium wilfordii polyglycoside suspension in deionized water was administered intragastrically,with the concentration being 5mg of Tripterygium wilfordii poly glycoside for every 1ml of said suspension,approximately 40 times the human dosage.Mice in all 4 groups(model group,control group,low TCM dosage group,high TCM dosage group)were administered 8ml/kg/d of Tripteiygium wilfordii polyglycoside suspension intragastrically at 10am for 30 days.Once the model mice were prepared,the following dosage regimen was administered intragastrically.Nonmal group:8ml/kg/d of deionized water at 10am for 30 days;model group:8ml/kg/d of deionized water at 10am for 30 days;control group:8ml/kg/d of L-Carnitine oral solution at 10am for 30 days;high TCM dosage group: 2.70g/kg/d of Yishen Jianpi formula at 10am for 30 days;low TCM dosage group:1.35g/kg/d of Yishen Jianpi formula at 10am for 30 days.During the course of the experiment,a weekly weighing of the mice was done to adjust the dosage according to increment in weight of each mouse by way of pharmacological experimental method calculations.After 30 days of medical intervention,the experimental mice were euthanized and their semen was collected and analyzed using specialized equipment to determine the motility of the sperm in each sample.Flow cytometry and fluorescent staining methods were used to detect changes in both the mitochondrial transmembrane potential as well as mitochondrial permeability transition pore.RESULTS:1 semen quality of miceThe percentage of grade A,A+B,PR+NP as well as sperm concentration in the control group,low TCM dosage group and high TCM dosage group was significantly higher than that compared to the model group(p<0.05).The percentage of grade A,A+B,PR+NP as well as sperm concentration in both TCM groups were significantly higher than that compared to the control group(p<0.05).The percentage of grade A,A+B,PR+NP as well as sperm concentration in both TCM groups and the control group were significantly different compared to the normal group(p<0.05).There was no significant difference between the low TCM dosage group and high TCM dosage group.2 mitochondrial membrane potential of miceThere was a significant decrease in the mitochondrial membrane potential in both TCM groups and control group compared with the normal group(p<0.05).There was a significant increase in the mitochondrial membrane potential in both TCM groups and control group compared with the model group(P<0.05).There was a significant increase in the mitochondrial membrane potential in both TCM groups compared with the control group(p<0.05).There was a significant increase in the mitochondrial membrane potential in high TCM dosage group compared to the low TCM dosage group(p<0.05).3 mitochondrial permeability transition pore of miceThere was a significant decrease in the mitochondrial permeability transition pore in both TCM groups and control group compared with the normal group(p<0.05).There was a significant increase in the mitochondrial permeability transition pore in both TCM groups and control group compared with the model group(P<0.05).There was a significant increase in the mitochondrial permeability transition pore in both TCM groups compared with the control group(p<0.05).There was a significant increase in the mitochondrial permeability transition pore in high TCM dosage group compared to the low TCM dosage group(p<0.05).CONCLUSION:1 Yishen Jianpi formula can decrease the damage dealt to mice sperm mitochondrial membrane potential from tripterygium wilfordii polyglycosides thereby increasing the mitochondrial membrane potential of mice sperm.2 Yishen Jianpi formula can decrease the damage dealt to mice sperm mitochondrial permeability transition pore from tripterygium wilfordii polyglycosides thereby improving the permeability of the mitochondrial permeability transition pore of mice sperm.3 Yishen Jianpi formula can decrease the damage dealt to mice sperm motility from tripterygium wilfordii polyglycosides thereby increasing the quality of mice semen.Part two:The clinical study on the treatment of oligospermia by Yishen Jianpi RecipeOBJECTIVE:This clinical research on clinical male sperm less weak as the research object,through the test before and after treatment all indexes to improve the effect of semen routine,to observe the men on the spleen and kidney party less weak sperm disease treatment effect.From the clinical perspective,the therapeutic effect of yishen jianpi prescription was observed and combined with experimental research to pro-vide a new way for the treatment of male infertility in the future.METHOD:The clinical research adopts self control study,before and after the observation period is 90 days,number of cases observed for 30 cases,drugs for international dongzhimen hospital pharmacy benefit spleen kidney drug particles,in the treatment of patients before and after each A semen routine testing,statistical analysis of semen routine of class A sperm vigour,(A+B)grade sperm vitality,sperm vitality,sperm concentration index.Every 30 days,all patients were followed up and asked about their spouse’s pregnancy,and evaluated the clinical efficacy.RESULTS:After treatment,to all the participants in the study were 30 patients semen routine testing,through the analysis,after treatment in patients with grade A sperm vitality,(A + B)grade sperm vitality,sperm vitality and sperm concentration was significantly improve,the difference is statistically significant(P<0.01);This study of the communist party of China to collect cases,32 cases according to the clinical curative effect evaluation standard,heal in 5 cases(15.6%),8 cases(25.0%),13 cases(40.6%),effective invalid 6 cases(18.8%),the total effective rate was(81.3%).CONCLUSION:YishenJianpi Recipe can obviously improve the semen quality of patients with deficiency of deficiency of spleen and kidney.
