Irritable bowel syndrome(irritable bowel syndrome)is a common disorder of gastrointestinal function,with abdominal pain,diarrhea,defecation habits and abnormality of defecation as the main symptoms,persistent or intermittent episodes,without the symptoms of morphological and biochemical abnormalities.According to the Rome III diagnostic standard,IBS can be divided into diarrhea(IBS-D),constipation(IBS-C),mixed type(IBS-M)and uncertainty(IBS-U),and IBS-D is the most common.IBS is very common in clinical practice.It is also widespread in the world.The prevalence rate can reach 8.8%.The incidence in Beijing is 7.26%.IBS is characterized by a long course of disease and recurrent seizures.It accounts for more than half of the number of outpatients in the digestive clinic.It brings great economic and mental burden to the society and family.The study found that mental state,social environment and life stress stimulation can influence the occurrence and development of IBS to some extent.Valeriana jatamansi is the dried roots and rhizomes of Valeriana jatamansi Jones.Originally recorded in Compendium of Materia Medica,it was collected in the 2015 edition of the People's Republic of China Pharmacopoeia.Its main functions are regulate the flow of vital energy and remove obstruction toit and relieving pain,eliminating food and stopping diarrhea,dispelling wind and removing dampness,and tranquilizing and allaying.excitement.Clinically,it can treat abdominal distension,abdominal pain,diarrhea,dysentery,wind-cold-wetness bype of arthralgia,soreness and weakness of waist and knees,insomnia and so on.The previous study found that the effective parts of valeriana jatamansi and iridoid can improve the gastrointestinal hyperactivity,visceral hypersensitivity and psychosis in IBS model rats induced by chronic stress.Previous studies showed that valeriana jatamansi could increase the score of horizontal movement and vertical movement of rats,and increase the sucrose preference of rats.valerians jatamansi can also play a significant therapeutic effect on visceral sensitivity,defecation and mental activity of IBS rats induced by slow acute stress.In addition,the effective fraction of iridoid can regulate the contents of peripheral and central 5-HT,and improve the gastrointestinal symptoms of IBS rats.Objective:In order to simulate a IBS animal model more suitable for clinical pathogenesis,this topic simulates IBS rat model with binding,ice water and tail stimulation on the basis of separation of mother and baby,In order to further investigate the effect of valeriana jatamansi fragrance extract on the efficacy of IBS and the regulation mechanism of 5-HT pathway,this topic is evaluated by observing the visceral sensitivity,fecal score,number of defecation particles,changes of mental activity and changes of intestinal flora in IBS model rats induced by three kinds of stress induced by valeriana jatamansi fragrance.Effect of valeriana jatamansi fragrance extract on the efficacy of IBS;By measuring the 5-serotonin(5-HT)in the cortex,hypothalamus,serum and colon of the rat model of IBS,the central and peripheral mechanisms of the effect of spider scent extract were preliminary explored.Methods:1.the IBS rat model was established by the method of mother to child separation and three kinds of stress:1 day male young male rats were randomly divided into 7 groups,with 12 groups in each group:the control group:no intervention.Mother to child separation combined with three stress groups:the newly born 2 days SD young rats were separated from the female rats,3 hours a day,20 days continuously,and then normal to 49 days,and fiftieth days to fix the SD rats with medical tape to fix their front and hind limbs,so that the rats could not move freely,3 hours a day.Fifty-first D SD rats were put in the bucket of ice water(4 degrees Celsius)and removed after 5 min.Fifty-second day SD rats were clamped by the hemostatic forceps(wrapped in cotton)to the tail of the rat,3 cm away from the root of the tail,and 1 min at the end of the clip.The above three stress methods,three days for a cycle,until ninetieth day.The high,medium and low dose group(36.9mg/kg,18.4mg/kg,9.2mg/kg)in the bacillus licheniformis group(150mg/kg)and the group of pinaverium bromide(20mg/kg)were in the same model group.After the success of the model,the dose group and the positive drug group began to fill the stomach every day on the ninety-first day.The blank control group and the model group were given the distilled water of the stomach and given the medicine for 35 days.The changes of abdominal retracement score,stool score,defecation grain number,body weight growth rate and intestinal flora abundance were used to evaluate the visceral sensitivity and gastrointestinal function changes of IBS model rats,and the changes of mental activity in IBS model rats were evaluated by the horizontal movement score and the vertical movement score in the open box test.2.by measuring the AWR score,the stool score,the number of defecation particles,the weight growth rate and the horizontal movement score,the vertical movement score and the intestinal flora abundance,the effect of the spider fragrance extract on the IBS model rats was explored.The AWR score of the experimental rats was measured in the 40,60,88,110 and 125 days of the experiment.On the 40,60,88,110,and 125 days of the experiment,the rats'fecal score was measured,and the weight growth rate of experimental rats was measured on the 42,57,71,87,93,108,123 days of the experiment,and the level of horizontal movement,the vertical movement score and the number of defecation particles were measured on the day 35,61,83,105 and 120 days of the experiment.Faeces of rats were used to detect the abundance and structural changes of intestinal flora in rats.3.the expression of 5-HT and 5-HIAA in cortex,hypothalamus,serum and colon were measured by high performance liquid chromatography.Results:1.before model building,there was no significant difference in AWR score,stool score,number of defecation particles,weight growth rate,horizontal exercise score and vertical movement score(P>0.05).After model building,the AWR score,stool score and defecation grain number of the model group were significantly higher than that of the blank control group(P<0.05),the weight growth rate and the horizontal transport rate.The scores of moving scores and vertical movements were significantly lower than those of the blank control group(P<0.05).2.after the drug of spider incense,the AWR score,fecal score and the number of defecation grains were significantly lower than the model group(P<0.05)in the group of high,middle and low dose of the spider incense extract,and the weight growth rate of the high dose group and the horizontal exercise score and vertical movement were significantly higher than the model group.Group(P<0.05).3.after treatment with spider extract,the abundance of intestinal flora in each treatment group also changed significantly.The distribution of intestinal flora in each group was similar,and the abundance of Bifidobacterium and Clostridium could be observed by the treatment of spider extract.The increase in the abundance of Bifidobacteria in the intestinal tract can enhance the intestinal defensive action mediated by epithelial cells,and reduce the content of 5-HT in the body,and improve the symptoms of diarrhea in IBS.The increase of Clostridium abundance can play a role in nutrition epithelium and tissue repair.4.The expression of 5-HT and 5-HIAA in the cortex,hypothalamus,serum and colon of the 4.model rats increased significantly(P<0.05).The high dose group of the spider incense extract could reduce the content of 5-HT and 5-HIAA in the cortex,hypothalamus,serum and colon(P<0.05).Conclusion:The IBS rat model can be successfully established by the combination of mother and infant separation and three kinds of stress.The spider extract has the effect of improving the visceral sensibility,hyperfunction of gastrointestinal function,abnormal mental activity and intestinal flora disorder in IBS model rats induced by maternal and infant separation combined with three kinds of stress.The mechanism may play a role in relieving the clinical symptoms of IBS by regulating the content of 5-HT and 5-HIAA in the central and peripheral regions.Through the study of this subject,it provides an experimental basis for the study of the material basis and mechanism of the IBS in the treatment of spider fragrance,and provides a new way of thinking for the study of the new clinical medicine for the treatment of IBS.
