Polyporus umbellatus is the dry sclerotium of Polyporaceae fungus Polyporus umbellatus(Pers.)Fries.It can promote diuresis and remove dampness and as a traditional Chinese medicine for treatment of edema,scanty urine,vaginal discharge,jaundice and diarrhea.In recent years,due to life pace quicking,polluted environment suffering,work pressures increasing,and corticosteroids and chemotherapy drugs long-term using,more and more people may cause immunity declined and the risks of infection increased.The polysaccharides of Polyporus(PPS)has obvious effect on immunoregulation,which is a water-soluble polysaccharide extracted from Polyporus.It has many pharmacological activities,such as anti-tumor,immune regulation,liver protection,anti-aging.It also has good therapeutic effects on malignant tumor and chronic hepatitis B.Many studies have shown that PPS can regulate immunity and scavenge free radical in vitro,but its immune function and antioxidant activity in vivo had not reported comprehensively.Therefore,in this experiment,PPS was extracted and purified by water extraction and alcohol precipitation method,and used to cruve the immunosuppressed mice model was established by cyclophosphamide(Cy)treatment.The objective of this study is researching the effects of PPS on immune organs,immune cells,immune factors and antioxidant activity in immunosuppressed mice,which can provide theoretical basis for further development and utilization of PPS.The main research contents and results are as follows:'1.PPS was extracted by water extraction and alcohol precipitation method,and the concentration of PPS was determined by phenol-sulfuric acid method.The results showed that it is available 2.34 g crude extract of PPS from 100 g dry Polyporus powder by alcohol precipitation method.The content of PPS from crude extract was 68.10%determined by phenol-sulfuric acid method.2.-To investigate the effects of PPS on immune regulation,72 Balb/c mices were randomly divided into normal group,Cy model group,positive group and three treatment groups with different doses of PPS(50,100,200mg/kg).On the 1?3 day,mice in Cy model group,positive group and treatment group with 80 mg/kg Cy solution through intraperitoneal injection to establish immunosuppressed mice model,while the mice in normal group were injected by normal saline.Thereafter,the mice in normal group and Cy model group were gavaged with 0.3 mL/d normal saline,mice in positive group were gavaged with 20 mg/kg levamisole hydrochloride(LH)and mice in the three groups were treated with PPS for 14 days.On the last day,besides the normal group,mice in the other five groups were administered Cy solution through intraperitoneal injection.After 24 hours,the mice were weighed and then were sacrificed via cervical dislocation,the blood and tissues samples were collected to measure the immune indexes.The results showed that Cy can decrease the weight of mice.According to the immune organ index,PPS can improve the thymus index and spleen index,but it has few effect on liver index.In addition,200 mg/kg PPS could significantly increase the content of serum protein and the number of peripheral blood cells(P<0.05).At the same time,PPS can improve spleen lymphocyte proliferation and the phagocytic function of peritoneal macrophages and increase the levels of NO,TNF-a and IL-1?.The results showed that 200 mg/kg PPS can obviously restore the immune function of immune injured mice and regulate the immune response of the immune system.3.The oxidative stress indexes SOD,MDA,T-AOC,CAT,GSH-Px were measured in mice liver.The results showed that Cy can significantly decrease the SOD,GSH-Px,CAT and T-AOC activity while increase the content of MDA(P<0.01)in liver tissues of normal mice.However the PPS can significantly improve SOD,GSH-Px,CAT,T-AOC activity,decrease the content of MDA(P<0.05)in liver tissues of immunosuppressed mice,suggesting that PPS can significantly reduce oxidative stress injury in the body and improve antioxidant capacity.From the above,PPS can enhance immunity by promoting immune responses and raising antioxidant capacity of immunosuppressed mice.
