Purpose:gut microbiota play an important role in the physiological process including the intestinal nutrient absorption,immune system development and regeneration of gastrointestinal epithelial cells,and established a close symbiotic relationship with the host WA is one of oldest,the most representative pupolation of China,the features of which settlement were shared centrally,mostly distribute in ximeng,and cangyuan,and menglian,and still remain a traditional and unitary diet.however,Han is the bigest population in China,which the indivial can be seen all aroud China even the world,and its diet is becoming more and more pluralistic by the shock of culture communication.At present,it is a international perception that diet,genetics and age and other factors had implications for the structure and composition of gut microbiota.This paper aims to reveal Han and WA's structure and composition of intestinal microflora in healthy subjects.and probes the variation of the gut microbiota with Han and WA populations,the effecting factors.Methods:Using 454 pyrosequencing,we analysed the gut bacterial communities and variations of 36 healthy subjects ranging in age from 1 to 95 years,including 25 Wa Chinese(nine families)from Yannan(Wa)and 11 Han Chinese with independent genetic backgrounds from Yannan(YN-Han),citing 7 Han Chinese(a family)from Hangzhou(HZ-Han)as control[1]Results:We obtained a high-quality 148,052 sequence data set(4112.5 ± 1147.7,SD,per sample)of the 16S rRNA gene V2 region by sequencing the 16S rRNA gene.A total of 3486 OTUs were identified according to 97%ID(545.861 ±155.858 OTUs,per sample).All identificated OTUs can be classified into 14 phyla,the two most dominant phyla was Firmicutes and Bacteroidetes.PCoA analysis results were shown to cluster by populaiton independently.the cluster concentration of Han-HZ and WA's were much better than Han-YN.We analyzed the structure and composition of gut microbitota of three groups at phylum and genera level and found that there were a significant differences among the three populations.We also compared the intestinal microflora of 25 WA samples,the results showed that there were similar at the phyla and genera level,there was a differences in abundances.Clustering analysis of 25 WA showed that there were three pairs cluster by couple,three pairs cluster by parents and children,and three sample cluster by age,indicating that these a high similar structure of intestinal flora were existed among those individuals.Then the comparison of the gut microbiota structure and composition among the father,mother and children from different families found that besides the father's group and child group has a significant differences in abundances of Actinobacteria,the others at phyla and genera level though has differences,but the difference was not significant.Besides,the prevolla abundance was found a much higher abundance in the man than the other three member within a samefamily.Conclusions:Taking Chinese as the big genetic background,we considerd that the three populations's genetic background has consistency.Based on the PCoA analysis results,we considered that differences on diet habits(especially pepper alkali of intake)was the main reasons resulting in the diversity of the Han-HZ and WA,and the similar of those two groups were derived from the same big genetic background.Besides,Han-YN volunteers were chosen randomly,from different of life environment,meaning that the gut microbiota were effect by interaction of many social factors such as diet.environment and life style an so on.The results from analysis of 25 WA samples showed that similar structure of intestinal flora were existed among those individuals,however,there was a differences,in abundances.Based on the results of cluster analysis,we believe that the same diet,genetics and age may increase similarity of the structure and composition of gut microbiota among couples,parents and children individuals.we considered that the interaction of distinct diet and genetic may play an important role in resulting in the variation of the gut microbiota between and within WA families.
