The research object of this paper is dried and mature seeds of Litchi chinensis Sonn.As a kind of natural Chinese medicine, the litchi seeds is made by plucking its ripe fruits in the summer, removing the skins and the fleshy ones, washing and drying. In this paper, the experiments and statistical methods are combined to provide a new scientific and effective method for quality evaluation of litchi seeds.First of all,the experiment was conducted by using the cold immersion ultrasonic extraction method.By 95% ethanol to extract 10 batches of litchi seeds from different sources.litchi seeds extracts were gradient separation by the macroporous resin column, then DPPH radical scavenging method was used to filter out the antioxidant activity fractions. The active fraction of litchi seeds were determined by DPPH and FRAP,and the total flavonoids content and total phenol content were also measured. The chemcial fingerprint of 10 batches of litchi seeds were established by HPLC.Then,15 common peaks were find out,and the spectrum relationship between the fingerprint and antioxidant activity was analyzed by multiple correlation analysis.Five active peaks were deduced according to the results of EC50 and FRAP partial correlation analysis.Combin TLC-bioautography assay with fingerprint of HPLC,the four active components of the litchi seeds were verified. In the end,the quality evaluation of litchi seeds from 10 different sources was evaluated by similarity analysis and cluster analysis(HCA),the results showed that batches with similarities less than 0.9 had weaker antioxidant activity and they were classified into one class in cluster analysis. The results show that this method is reliable for the quality evaluation of litchi seeds, and also provides a new way for the research of other traditional Chinese medicine.
