| Purpose:To observe the effect of electroacupuncture on the expression of cholesterol 7α-hydroxylase(CYP7A1)in the liver in atherosclerosis rabbit,to explore the new mechanism of acupuncture in the treatment of atherosclerosis,and to study the therapeutic effect of acupuncture on atherosclerosis and related diseases Provide experimental basis.Material and method:The 26 male New Zealand rabbits,weighing about 2.5kg ±0.5kg,aged from 2 to 3 months,were divided into cages and fed for a period of 1 weeks.The rats were randomly selected as the blank group,and the remaining 19 were the model group.Control group fed with normal diet,model group fed with high-fat diet.After 4 weeks of high fat diet,model group rabbits underwent carotid balloon injury surgery,postoperative high fat diet for 4 weeks,from the blank control group and model group randomly selected rabbit carotid artery pathological section observed by HE carotid plaque morphology staining to determine the model is successful or not.After successful modeling,18 rabbits were randomly divided into three groups: model group,electro acupuncture group and Western medicine group(n = 6)in each group(n = 3).The model group was not given any treatment;the western medicine group Atorvastatin Calcium Tablets suspension gavage;Electroacupuncture group Neiguan,Zusanli and Guanyuan for acupuncture treatment,1 times a day,each time for 20 min,6 for 1 courses,1 days rest between treatments,a total of 4 courses.After the treatment,blood samples were collected from the ear vein of rabbits,the blood lipid levels of each group were detected,and the pathological changes of carotid atherosclerotic plaques were observed by HE staining;The expression of CYP7A1 protein in rabbit liver was detected by Western blot method.The expression of CYP7A1 m RNA in rabbit liver was detected by RT-PCR.Results: 1.Blood lipid levels: from control group,model group,TG and CHO in rabbit blood LDL content increased,HDL content decreased,with statistical significance(P<0.01),compared with the model group,electro acupuncture group,western medicine group,TG and CHO in rabbit blood LDL content were significantly decreased,HDL content increased,there statistical significance(P<0.01),EA group compared with the western medicine group,no significant difference(P>0.05).2.The expression of CYP7A1 protein:Compared with the blank group,the expression level of CYP7A1 protein in the model group decreased,with statistical significance(P<0.01);Compared with the model group,electroacupuncture group and Western medicine group,the expression level of CYP7A1 protein in rabbits increased significantly,with statistical significance(P<0.01);Electroacupuncture group and Western medicine group was no significant difference between groups(P>0.05).3.The expression of CYP7A1 m RNA: Compared with the blank group,the expression level of CYP7A1 m RNA in the model group decreased,with statistical significance(P < 0.01);Compared with the model group,the expression level of the electroacupuncture group and medicine group in CYP7A1 m RNA was significantly increased,with statistical significance(P<0.01),electro acupuncture group and Western medicine group was no significant difference between groups(P>0.05).Conclusion: 1.Electroacupuncture at Neiguan,Guan Yuan and Zusanli can improve the blood lipid level of atherosclerotic rabbits.2.Electroacupuncture at Neiguan,Guan Yuan and Zusanli can promote the expression of CYP7A1 m RNA and protein in the liver of rabbits,which may be one of the mechanisms of electroacupuncture treatment of atherosclerosis. |
