make a difference of Blackberrylily Rhizome.and its concocted products.Discuss the purpose of Blackberrylily Rhizome.with violet bamboothe and TCM for Blackberrylily Rhizome transformation provided the scientific basis of medicine.Material and method: 1.Compared the total flavonoids of products with processed products of Blackberrylily Rhizome by UV spectrophotometry.It is aimed at compareing the effects on total flavonoids content of products with concocted products and analysising the difference.2.Using ultra-high performance liquid chromatography system.Chromatographic conditions,Column:ACQUITY UPLC ?BEH C18(2.1mm×50mm,1.7?m);Mobile phase: 0.1% phosphoric acid aqueous solution(A)-acetonitrile(B)-0.1% phosphoric acid aqueous solution containing 0.5% methyl-?-cyclodextrin(C),by gradient elution;Flow rate:0.2m L·min-1,column temperature is 40?,detection wavelength is 265 nm.Determine the contents of tectoridin,iridin,tectorigenin,iristectorigenin A,Iristectorigenin B,Irisflorentinand,analysis the difference of contents.3.Male rats were randomly divided into blank group,model group,positive drug group,the control group of raw materials and dry hair dry system,the use of gavage mode,after 7 days continuous administration,1% carrageenan was injected into the right toe of the rats at 30 min after the last administration.The foot volume of the rat was measured at the same time,and the toe swelling rate was calculated.SPSS statistical software was used to analyze the inhibitory effect of raw materials and processed products on the swelling of the toes in rats. 4.The rats were randomly divided into three groups and sacrificed for 7 days.The rats were gavaged with products,concocted products and the volume of physiological saline.Serum sample after the last delivery,the determination of serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)levels,explore the Blackberrylily Rhizome raw product and concocted products of its influence on blood biochemical indexes.5.Choose Beagle dogs as experimental animals,a single gavage is given to the product.Determine the concentration of Tectorigenin and Iris in serum at different time points in Beagle dogs by HPLC-MS.Calculate pharmacokinetic parameters and analysis by DAS.2.0.And make a comparison with previous research and pharmacokinetic parameters of Blackberrylily Rhizome raw product.Results: 1.The results show that total flavonoids content compared with Blackberrylily Rhizome raw product,the total content of flavonoids in the Blackberrylily Rhizome processed products reduced 9.89%.2.The results of the content of monomer active component showed that the content of tectoridin,iridin,tectorigenin,iristectorigenin A,iristectorigenin B,irisflorentin irisflorentin and Dichotomitin in raw products are 5.849mg·g-1,16.74mg·g-1,0.9770mg·g-1,1.040mg·g-1,0.4290 mg ·g-1,6.992mg·g-1,1.433mg·g-1,0.3392mg·g-1 and which in processed one are 3.102mg·g-1,7.801 mg·g-1,0.8385mg·g-1,0.7862mg·g-1,0.3221mg·g-1,4.595mg·g-1,1.175mg·g-1,0.2725mg·g-1.Comp ared with raw products,the 8 active components are all decline.The values of declining are 46.96%,53.40%,15.90%,24.40%,24.92%,34.28%,18.00% and 19.66%.there is a new peak appears in the 8min retention time in the concocted product,which indicates that processed with bamboo can change the chemical composition of Blackberrylily Rhizome.3.Experimental results of rat foot swelling showed that the swelling of the rat toes was significantly inhibited(P<0.05,P<0.01)in both products and concocted products compared with the model control group,suggest that raw materials and dry products are in vivo anti-inflammatory effect;It suggested that processed products couldn't make the anti-inflammation activity stronger(P>0.05).4.The experiment results show that contents of ALT and AST in the blank group are 24.34±10.0U·L-1,26.41±7.10U·L-1;The content of ALT and AST of raw products respectively37.96±17.2U·L-1,42.91±18.4U·L-1;The content of ALT and AST of processed products are 23.11±13.2U·L-1;27.74±13.8U·L-1.Compared with the blank group,there are more AST and ALT in the rat serum,the dofference is ststistically significant(P<0.05),which show that Belamcanda chinecsis rhizoma is helpful to improve AST and ALT;The contents of AST and ALT in the processing group is similar to the blank group,it has no ststistically significant(P>0.05).The contents of AST and ALT in processing drugs are markedly lower than Belamcanda chinecsis rhizoma,the dofference is ststistically significant(P<0.05),which show that the processing drugs are helpful to reduce AST and ALT.5.The pharmacokinetic experiment results show that the LC-MS is adopted to establish the products during the tectoridin and tectorigenin analysis method,separate the two ingredients well,detection range from 0.0977ng·m L-1to31.25ng·m L-1and0.1114ng·m L-1 to 35.62ng·m L-1,Low,medium and high three levels between batch and batch of precision,accuracy,and the matrix effect meets the requirements.2 kinds of components in serum of pharmacokinetic model for the second chamber,best products the main pharmacokinetic parameters for blackberry lily: The Tmax(h)of tectoridin is1.5±0.62,the Cmax(ng·m L-1)of tectoridin is 5.678±3.723,The Tmax(h)of tectorigenin in processed products is 3.25±1.38,the Cmax(ng·m L-1)of tectorigenin in processed products is16.83±2.78,Compared with the results of the previous study by gavage beagle with Belamcanda chinecsis rhizoma,The Tmax(h)of Tectoridin and tectorigenin in processing drugs and Belamcanda chinecsis rhizoma,which shows that it has no ststistically significant(P>0.05),but the Cmax within the two groups shows that the defference is ststistically significant(P<0.05),and the Cmax of Tectoridin in processing groups are lower than Belamcanda chinecsis rhizoma groups,the Cmax of tectorigenin in processing groups is lower than Belamcanda chinecsis rhizoma groups.Conclusion: By analysising the chemical composition,anti-inflammatory activity in the body and blood biochemical indexes AST,AST test results of Blackberrylily Rhizome before and after processing,indicated processed products have no obvious effect,but it can reduce the damage of rats to liver and spleen,suggest that Blackberrylily Rhizome processed by violet bamboo is a kind of processing method of given priority to attenuation.
