Purpose:To study the effect of Dianxianqing Particles on the expression of p-tau in brains and Aβ production pathway in mice induced by Aβ1-42.Material and method:The mice were randomly divided into sham operation group,model group,donepezil 1.3mg / kg control group,Dianxianqing Particles 12.48 g / kg group,Dianxianqing Particles 6.24 g / kg group,Dianxianqing Particles 3.12g/kg group.The mice were injected with 3μl condensed Aβ1-42(1 mg/ml)into the lateral ventricle.The rats were dosed for the next day for 21 days.The Y-maze behavioral test was performed one hour after the end of the 15-day administration,and then the morris water maze behavior test was performed for 6 consecutive days.,once a day.The 15 th day of the Y maze test,the first 16 d to 21 d water maze behavior test.To investigate the effect of epilepsy granules on the behavior of AD mice.The expression of tau-5,Ser 404,Thr 181,GSK-3 Beta,phospho-GSK-3 in the brain of AD mice were observed by HE staining.The expression of tau-5,Ser 404,Thr 181,GSK-3β,Akt,phospho-Akt,PI3 K and phospho-PI3 K were used to investigate the effect of Dianxianqing Particles on the phosphorylation of Tau protein in AD mice.Immunohistochemical staining was used to observe the expression of Aβ1-42,Aβ1-40 The expression of Aβ1-42 and Aβ1-40 in brain tissue of AD mice was detected by ELISA.The expression of BACE 1 and PS 1 was detected by Western blot.The effects of Dianxianqing Particles on AD model mouse Aβ productionResults: 1.Dianxianqing Particles can improve the learning and memory ability of AD model mice,and have protective effect on neurons.2.Dianxianqing Particles can decrease the expression of Ser404 and Thr181 protein and inhibit the abnormal phosphorylation of Tau protein.3.Dianxianqing Particles can increase the protein expression of phospho-GSK-3 Beta and inhibit the activity of GSK-3β.4.Dianxianqing Particles can activate the brain PI3 K / AKT pathway,inhibition of Tau abnormal phosphorylation.5.Dianxianqing Particles can reduce the deposition of Aβ1-42 and Aβ1-40 in brain tissue of AD mice,and decrease the content of Aβ1-42 and Aβ1-40 in brain tissue of AD mice.6.Dianxianqing Particles can reduce the activity of BACE 1,PS 1,thereby reducing the production of AβConclusion: 1.Dianxianqing Particles can reduce Aβ1-42 induced neuronal damage,thereby improving AD model mice learning and memory ability;2.Dianxianqing Particles can inhibit the abnormal phosphorylation of Tau protein,thereby delaying the course of AD.3.Dianxianqing Particles can reduce the content of Aβ in brain of AD mice by inhibiting the activity of BACE 1 and PS 1,and reduce their damage to brain tissue...
