| Purpose: Observe the effect of atherosclerosis model rabbits by electro-acupuncture at Neiguan,Guanyuan and Zusanli.Explore the relationship between electro-acupuncture at Neiguan,Guanyuan,Zusanli and the inhibitory of inflammatory response of atherosclerosis model rabbits by detecting the level of MIF and IL-10 in the serum.Material and method:Ordinary male New Zealand rabbits 26,body weight 2.0-2.5kg,2-3 months.Establish the AS model by high fat diet and combined carotid artery balloon injury.Divide the rabbits into two groups randomly: control group(n = 7)and model group(n = 19).The rabbits of control group are fed with normal diet every day.The rabbits of model group are fed with normal diet one day and high-fat diet the next day.After four weeks,carotid artery balloon injury is applied to the rabbits of model group.Then go on feeding for 4 weeks.After eight weeks,kill one rabbit from each group to confirm the atherosclerotic plaques by HE staining.After modeling,feed the control group(6 rabbits)as before.Divide the rabbits of model group into three groups: AS model group(6 rabbits),electro-acupuncture group(6 rabbits),positive drug control group(6 rabbits),feed with normal diet one day and high-fat diet the next day,and start treatment.Six day for a course of treatment,1 day rest between the two courses,continuous 4 courses.Therapeutic method: electro-acupuncture group :acupuncture "Guan Yuan,Neiguan,Zusanli" point of treatment,after the needle,connecte the electro-acupuncture treatment instrument to the "Neiguan,Zusanli" acupuncture points,with dense waves,continue 20 minutes,1 times / day;positive control group: atorvastatin calcium tablets 1mg / kg / d gavage treatment.At the end of the fourth course,collect the blood samples from the ear of the experimental rabbits.The pathological changes of atherosclerotic plaques were observed by HE staining.Detect the levels of MIF and IL-10 in rabbit serum.Observe the effect of inflammatory response by acupuncture at Guanyuan,Neiguan and Zusanli points,to explore the mechanism of electro-acupuncture stimulation of Neiguan,Guanyuan and Zusanli points on atherosclerosis.Results: 1.Under light microscope: The rabbit 's aortic intima was completely continuous in the control group,none inflammatory cell infiltration and foam cell formation.In the AS model group,the intima of the rabbit aorta are significantly thickened,the endothelial cells are discontinuous,monocytes,macrophages are infiltrated and aggregated,and a large number of foam cells are clustered into patches.Electroporation and positive drug control group,the rabbit aortic intima and wall thickening,endothelial cells are not continuous,showing mononuclear cells,macrophage infiltration,foam cell formation,lesions than the AS model group was significantly reduced.2.Compared with AS model group,both electro-acupuncture and positive drug intervention can down-regulate the content of MIF in serum of AS rabbits.(P <0.05).There is no significant difference between the electro-acupuncture group and the positive control group(P> 0.05)about the level of MIF.3.Compared with AS model group,electro-acupuncture and positive drug intervention can up-regulate the content of IL-10 in serum of AS rabbits.(P <0.05).There is no significant difference in serum IL-10 level between the electro-acupuncture group and the positive control group(P> 0.05).Conclusion: 1.Electro-acupuncture Guanyuan,Neiguan,Zusanli points,can improve the atherosclerotic plaque situation,delay the further development of atherosclerosis.2.Electro-acupuncture Guanyuan,Neiguan,Zusanli points,can restrain the secretion proinflammatory cytokine MIF,and promote the expression of anti-inflammatory factor IL-10 in serum,and play an anti-inflammatory effect in the treatment of atherosclerosis. |
