Purpose:To establish a model of Candida albicans infection in splenic mice and observe the weight and ultrastructural changes of small intestinal epithelial cells before and after infection.To investigate the effect of Grifola frondosa polysaccharide on the carrier of Candida albicans and the expression of IL-1? in the small intestine.For the clinical treatment of Candida albicans infection in the intestine of the spleen,the use of traditional Chinese medicine-Grifola frondosa polysaccharide treatment,to provide a reliable experimental basis.Material and method:SPF grade Kunming mice were divided into normal control group(A),spleen deficiency model group(B),spleen deficiency+Candida albicans(C),spleen deficiency+Candida albicans+Fluconazole group(D),spleen deficiency+Candida albicans+Grifola frondosa polysaccharide group(E).After one week of sacrifice,the spleen deficiency model was started according to the reference.Except for group A(the normal control group),The model of B,C,D,E group were established by swimming,feeding cabbage and lard,and other compound factor.Single day swimming group,swimming until the endurance were exhausted,then fed cabbage and free to drinking water,but fasting pellet feed;Two days model group was fed lard by oral lavage,each rat was given 0.2 m L/10 g body weight,then feeding pellets and free drinking water,maintain this method for 14 days.On the 15 th day of the experiment,in addition to group A and B,the remaining groups were given Candida albicans suspension with a concentration of 2×108 CFU/m L,each rat was given 0.2 m L/10 g body weight;A and B group were given the same amount of saline.On the 16 th day of the experiment,the mice in group D were orally treated with fluconazole solution;the mice in group E were orally treated with Grifola frondosa polysaccharide.The rats in each group were treated once a day for one week,the changes in the general state of the mice were observed.On the 15 nd and 22 nd day of the experiment,the mice were weighed and the feces was collected,the determination of the amount of bacteria in the manure was carried out;the jejunum tissues of each group were collected and scanned by transmission electron microscopy.The ultrastructural changes of small intestine mucosa were observed.The expression of cytokine IL-1? in intestinal tissue of mice was detected by enzyme-linked immunosorbent assay(ELISA).Results: 1.Effects of Grifola frondosa polysaccharides on the general state of Candida albicans after intestinal infection in spleen-deficiency mice Compared with the normal control group,the spleen deficiency model group,spleen deficiency+Candida albicans group began on the 15 th day of the experiment,showing the appetite reduction,dullness,weight loss,loose stools and other symptoms;One week after the trial drug treatment(on the 22 th day of the experiment),spleen deficiency+Candida albicans group mice diet was significantly reduced,weight decreased significantly,afraid of cold,like to snuggle together,limbs weakness and even walking sloshing performance.Compared with the spleen deficiency+Candida albicans group,the spleen deficiency+Candida albicans+Fluconazole group and the spleen+Candida albicans+Grifola frondosa polysaccharide group,the general state of the mice after the drug treatment have improved,there have been increased appetite,weight began to rise,fecal loose loose symptoms have improved.2.Effects of polysaccharides from Grifola frondosa on the number of staphylococcus faecalis after intestinal infection in patients with spleen deficiency Compared with the normal control group,the number of Candida albicans in the feces of the spleen deficiency model group increased slightly,but there was no significant difference between the two groups.Compared with the spleen deficiency model group,the number of Candida albicans in the feces after infection with Candida albicans was significantly increased(P<0.01).Compared with spleen deficiency+Candida albicans,the number of Candida albicans was significantly decreased(P<0.01)in spleen deficiency+Candida albicans + Fluconazole group,spleen deficiency + Candida albicans + Grifola frondosa polysaccharides in mice feces in the number of Candida albicans no significant difference.3.Effects of Grifola frondosa polysaccharides on body weight after Candida albicans Infection in spleen deficiency mice Compared with the normal control group,the average body weight of the spleen deficiency model group,the spleen deficiency+Candida albicans group,the spleen deficiency+Candida albicans+Fluconazole group,the spleen deficiency+Candida albicans+Grifola frondosa polysaccharide group were significantly decreased(P<0.01).Compared with the spleen deficiency model group,the mean body weight of the spleen deficiency+Candida albicans was significantly decreased(P<0.01).Compared with spleen deficiency+Candida albicans,the average body weight of the spleen deficiency + Candida albicans + Fluconazole group and the spleen deficiency + Candida albicans + Grifola frondosa polysaccharide were significantly increased(P<0.01).There was no significan difference between the spleen deficiency + Candida albicans + Fluconazole group and the spleen deficiency+Candida albicans+Grifola frondosa polysaccharide groups.4.Effects of polysaccharides from Grifola frondosa on the ultrastructure of small intestinal mucosa after intestinal infection of Candida albicans in spleen deficiency mice Compared with the normal control group,the microvilli of the small intestinal epithelium cells in the spleen deficiency model group were slightly shorter,but the arrangement of the cytoplasm was slightly decreased.After the spleen deficiency was infected with Candida albicans,the villi of the small intestine were sparse,The number and structure of organelles in the cytoplasm have different degrees of change,showing a vacuolar change.Compared with spleen deficiency+Candida albicans,spleen deficiency+Candida albicans+Fluconazole group,spleen deficiency+Candida albicans+Grifola frondosa polysaccharide group mice on the 7th day of drug intervention,the mouse small intestine villi and other structures are recovery,arranged neatly,the cytoplasmic organelles and the degree of abundance has also improved,occasionally changes in vacuoles.5.Effects of Grifola frondosa polysaccharides on IL-1? in intestinal tissue of Candida albicans after intestine infection in spleen deficiency mice Compared with the normal control group,the content of other mouse cytokine IL-1? in small intestine increased(P<0.05 or P<0.01);Compared with the spleen deficiency model group,the content of IL-1? in small intestine of mice infected with Candida albicans was significantly increased(P<0.01).Compared with spleen deficiency+Candida albicans,the levels of cytokines IL-1? in small intestine of mice with spleen deficiency+Candida albicans+Fluconazole and spleen deficiency+Candida albicans+Grifola frondosa polysaccharides group were significantly reduction(P<0.01).There was no significan difference between the spleen deficiency + Candida albicans + Fluconazole group and the spleen deficiency + Candida albicans+Grifola frondosa polysaccharide groups in IL-1?.Conclusions: 1.Grifola frondosa polysaccharide can improve the intestinal infection of Candida albicans splenic mice in the general state,increased spleen infection in Candida albicans mice weight and significantly reduced fecal bacteria.2.Grifola frondosa polysaccharide can reduce the pathological changes of small intestine mucosal ultrastructure in intestinal infection of Candida albicans spleen deficiency mice.3.Grifola frondosa polysaccharide can reduce the content of cytokines IL-1? in small intestine of mice with intestinal infection.
