The immunoassay is one of the current detection of beta-receptor agonists.The traditional immunoassay method use a strong specific antibody which can only recognize a single beta-receptor agonist.The key of multi-residue immunoassay is synthesis of the"common hapten" or "multi-cluster" artificial antigen,while the design of traditional hapten can only be according to experience.With the help of molecular modeling technique,this article design and synthesize "common hapten" and synthesize many kinds of "multi-cluster" artificial antigens,which provide material basis for detecting beta-agonists.This article is divided into four chapters,the main contents is as follow:(1)Summarize beta-agonists' development,pharmacological mechanisms,hazards,means of detection and outlining the research progress of multi-residue immunoassay.In addition,this chapter explain the topic basis of this article;(2)Build pharmacophore model of beta-agonists and design two "common hapten".Each compound's synthesis process is introduced in detail,and their structure characterization is carried out;(3)Use method of homology modeling to build the crystal structure of beta3-receptor and use method of molecular simulation to study the two beta agonist's "common hapten" and interaction mechanism with beta3-receptor,and calculate stable configuration and the lowest energy of the two beta agonist "common hapten".(4)Put several typical beta agonists of small molecules to the same BSA and made several "multi-cluster" and characterization and roughly calculate the number of coupling ratio.Results show that the process of small molecules coupling to BSA is successful and coupling ratio's range is better.At the same time,we put the artificial antigens for animal immunization and get artificial antibodies in serum and measure the valence of antibodies by indirect enzyme-linked immune,which appears that antibodies' valence of antibodies is well.
