Herba leonuri is the fresh or dry overgroud part of the labiate Leonurus japonicus Houtt,which can promot blood flow and regulate the menstrual function.Because the different areas have different climate and soil environment,herba leonuri has different harvest time and harvest organ,which cause the wide difference of drug quality,and affect the efficacy of herba leonuri.So,in response to the problems above,the experiment adopted hydroponic to culture herba leonuri in order to advance the quality and stablity of drug source.Hydroponics is an advanced plant cultivation technology,which with good commodity value and social benefit,can effectively shorten the plant growth cycle.At the same time,Hydroponics without regional soil limit,can eliminate regional differences,avoid the influence of light,water and temperature,reduce the pest damage in soil,and prevent the soil of continuous cropping obstacles.People can control the growth process in cultivation,in order to improve the production efficiency,and provide a reference for the hydroponic cultivation of Chinese herbal medicines in future.The experiment contents and results are as follows:1.The analysis of the effective components contents of herba leonuri pieces from eleven different areas found that there were significant difference for regional contents,Stachydrine hydrochloride contents varied at 0.1018%~2.1416%,and five of them didn’t reach the pharmacopoeia standards.Furthermore,it’s found that microelements contents of herba leonuri pieces varied greatly.2.Study on the adaptability of herba leonuri to hydroponics and the prescreening of nutrient solution formula.The result showed that herba leonuri can survive in nutrient solution,and its root structure changed to adapt to hydroponics environment.The roots must be prevented from light,and ventilated frequently in nutrient solution at low concentrate.Finally,hydroponics environment and measures were determined.By comparing the effects on different nutrient solutions as MS,1/2MS,Hoagland formula and 1/2 Hoagland formula,1/2 Hoagland formula was found the best one.3.The allocation of the proportion of macroelements in hydroponic nutrient solution was studied.Orthogonal design was used to screen out the optimal proportion of four macroelements in 1/2 Hoagland formula.The result showed that The herba leonuri fresh weight biomass was significantly related to KN03,and the content of stachydrine hydrochloride was significantly related to Ca(NO3)2·4H2O.Therefore,potassium nitrate and Ca(NO3)2·4H2O were adopted with 3/4 Hoagland formula,while others with 1/4 Hoagland formula according to the cost principle.The specific formula is that Ca(NO3)2·4H2O 708.75mg/L,KNO3 455.25 mg/L,NH4H2PO4 28.75 mg/L,MgSO4-7H20 123.25 mg/L.4.The allocation of the proportion of microelement in hydroponic nutrient solution was studied.The amount of Fe、Mn、Zn、Cu were adjusted based on general formula,and the Single factor design result showed that the best microelement formula was that H3BO3 2.86 mg/L,MnSO4·4H2O 2.13 mg/L,ZnSO4·7H2O 0.88 mg/L,CuSO4.5H2O 0.02 mg/L,:(NH4)6Mo7O24·4H20 0.02 mg/L,FeS04·7H2O-EDTA-2Na 6.59 mg/L-9.325 mg/L.
