GZ-107 is a new compound which derives from Glycyrrhetinic acid.To further evaluate the potential of GZ-107,the experiments on the GZ-107 anti-inflammatory effect and mechanism of LPS-induced murine macrophage inflammatory models were carried out to make a preliminary study.Elisa kit,RT-PCR experiments were used to determine the release of TNF-a,IL-1? and expressions of mRNA.We used Western Blot technique to measure the influence of GZ-107 in NF-?B and MAPK signaling pathway.The results showed that GZ-107 can inhibit the release of TNF-a and IL-1? and the expressions of their mRNA in a concentration-dependent manner.GZ-107 can block the entry of p65 into the nuclear and affect the phosphorylation level of I?B?IKK.GZ-107 can also inhibit the phosphorylation level of p38?JNK which was induced by LPS.This study demonstrated that GZ-107 can regulate the inflammatory process through influencing nuclear transcription factor NF-?B and activating signal protein kinase MAPK.Therefore GZ-107 may possess potential clinical value. |
