CIR(cerebral ischemia reperfusion)is a kind of ischemic cerebrovascular disease,which mainly refers to the brain damage caused by cerebral ischemia.,reperfusion blood flow,further aggravate the phenomenon of ischemic injury.CIR is a complex cascade of pathological process,involving many factors,including oxidative stress,inflammation,intracellular calcium overload,excitatory amino acid toxicity,blood-brain barrier damage,apoptosis and stress etc.Oxidative stress and inflammation are the two most important factors leading to CIR.The scientific name of rooibos is Aspalathus Linears,which is dry leaf and twig part of a leguminous plant(Barm.f.)Aspalathus linearis R.Dahlgren,a plant endemic to South Africa west area hampton.Research shows that rooibos has better antioxidant and anti-inflammatory properties,but for its role in CIR animal is not clear.This study intends to investigate rooibos effects on CIR mice,and to discuss its mechanism of action.Objective:Based on the previous research,this study intends to observe the rooibos effects on CIR mice,and to explore its possible mechanism.Methods:The ICR mice were randomly divided into control group,CIR group,rooibos low dose group,middle dose group and high dose group.Before the preparation of the model,each group was given drugs 7 days,and the control group and CIR group were given the same volume of distilled water,one time a day.After one week of administration,the model of middle cerebral artery occlusion(MCAO)was used to prepare the model.After the model was successfully established,the mice were administrated by the above method.1.The effects of rooibos on cerebral ischemia reperfusion injury of nerve function recovery(1)evaluate the success of model according to Longa neural function score method;(2)observe the cerebral infarction volume changes by TTC staining method;(3)the beam balance test(balance beam test)is to evaluate the motion coordination function,the rotating angle(corner test)is to evaluate the sensory function,water maze test(Morris water maze test)is to evaluate the cognitive function.2.The study of rooibos on cerebral ischemia reperfusion injury protection mechanism(1)Oxidative stress protective mechanism of rooibos on cerebral ischemia reperfusion in miceBy colorimetric method,determinate the damage indexes of oxidative stress in brain homogenates:superoxide dismutase(SOD)activity,catalase(CAT)activity and glutathione peroxidase(GSH-Px)activity,malondialdehyde(MDA)content,glutathione(GSH)content and oxidized glutathione(GSSG)content.(2)Anti-inflammatory protective mechanism of rooibos on cerebral ischemia reperfusion in miceBy enzyme-linked immunosorbent assay(ELISA),determinate the changes of inflammatory factors and related enzymes in brain homogenates:interleukin 1?(IL-1?)content,tumor necrosis factor a(TNF-a)content,intercellular adhesion molecule-1(ICAM-1)content and myeloperoxidase(MPO)activity.Results:1.Compared with the control group,the infarct volumes of CIR group were significantly increased(P<0.01)3 and the neurological function scores were significantly higher than that of the control group.The balance beam scores were significantly higher(P<0.01),the frequencies of turning to the damage side were significantly decreased(P<0.01),and the learning and memory decreased.Compared with the CIR group,the rooibos groups(P<0.05)significantly reduced the infarct volumes,neurological function improved slightly,the balance scores decreased significantly(P<0.01),the frequencies of turning to the damage side were significantly raised(P<0.01),and the learning and memory improved.2.After preparation of the model and administration drugs 7 days,compared with the control group,the activities of SOD,CAT in brain tissue of CIR group were significantly decreased(P<0.01)and MDA contents were significantly increased(P<0.05),GSH contents were significantly lower(P<0.01),GSSG contents increased significantly(P<0.01).Compared with the CIR group,the activities of SOD,CAT and GSH contents in brain tissue of rooibos groups increased significantly(P<0.05),the contents of MDA and GSSG were significantly lower(P<0.01 or P<0.05).After preparation of the model and administration drugs 14 days,compared with the control group,SOD activities in brain tissue of CIR group were significantly lower(P<0.01),MDA contents increased slightly,CAT activities decreased slightly and GSH-Px activities decreased slightly.Compared with the CIR group,SOD activities,CAT activities in brain tissue of rooibos groups increased slightly,MDA contents decreased slightly,GSH-Px activities increased slightly.3.Compared with the control group,the contents of TNF-?,ICAM-1 and the activities of MPO in brain tissue of CIR group significantly increased(P<0.05),the amounts of EL-1?increased slightly.Compared with the CIR group,the contents of TNF-?,IL-1?,ICAM-1 in brain tissue of rooibos groups decreased significantly(P<0.01 or P<0.05),MPO activities decreased slightly.Conclusions:1.Rooibos has a protective effect on brain tissue of CIR mice and can promote the recovery of neural function,the specific performance:significantly reduce infarct volumes and improve neurological function(nerve function score),coordinate body movement(balance beam test),improve the sensory function(rotation experiments),improve the ability of learning and memory(Morris water maze).2.Rooibos can alleviate oxidative stress,which is caused by cerebral ischemia reperfusion,the effect may be through improving the antioxidant system of brain function.3.Rooibos can alleviate inflammation which is caused by cerebral ischemia reperfusion,the effect may be related to reduce inflammation factors,inhibit the enzymes activities.In summary,rooibos has a protective effect on cerebral ischemia reperfusion injury,the effect may be related to its antioxidant and anti-inflammatory response.
