Effect Of Si Miao Yong An Decoction On Anastomotic Restenosis And VEGF Protein Expression After Vascular Anastomosis In Rats

Objective: To investigate the effect of Si Miao Yong An Decoction on anastomotic restenosis and expression of VEGF protein in microvascular anastomosis,and to provide theoretical support for clinical application.Methods: 27 rats,12 weeks old,weighing 220-250 g,SPF grade and male SD rats were used as the research object.Three were used for pre-experimenting.The rats were familiar with the tail artery anatomy and mastering the modeling technique.Male SD rats were randomly divided into two groups(n = 12):(1)the experimental group: after modeling to Si Miao Yong An Decoction Feeding;(2)the control group: after modeling to normal saline Feeding.On the 3rd day and the 7th day after the operation,the blood vessel tissue samples were collected at 2h after the last Feeding.each group were divided into two groups again.Vascular tissue specimens were fixed,embedded,sliced and other treatment,for HE staining observation of histomorphological changes and immunohistochemical detection of VEGF protein expression.Image analysis software IPP 6.0(Image-Pro-Plus 6.0)analysis of pictures,access to information with SPSS17.0 software for statistical data.Results: The experimental data were statistically analyzed.The difference of the intima area between the experimental group and the control group at 3d and 7 days after operation was statistically significant(P <0.05).The difference of PA / EELA ratio between the two groups at 3d and 7 days after operation was statistically significant(P <0.05).The difference of mean optical density between the two groups at 3d and 7 days after operation was statistically significant(P <0.05).Conclusion:(1)Si Miao Yong An Decoction can promote vascular endothelial cell replication and proliferation,speed up vascular repair,to achieve the role of anti-vascular anastomotic restenosis.(2)Si Miao Yong An Decoction can promote the expression of VEGF protein in the vascular anastomosis of rats,and play a biological effect on the endothelial cells.It is beneficial to the rapid repair of the intima and the formation of neovascularization.(3)Si Miao Yong An Decoction can anti-vascular anastomotic restenosis,improve vascular anastomotic patency rate.(4)Si Miao Yong An Decoction against vascular anastomotic restenosis of its role may be achieved through a variety of ways,the specific effect mechanism also need further experiments to explore the proof.
