Objective:This research is to discover the effect of dendrobium polysaccharides on rat liver fibrosis through the detection of serological indicator, pathology, collagen type I, collagen type III, Transforming growth factor-?1, MMP-13 and Tissue inhibitor of metalloproteinase-1, to reveal the role of dendrobium polysaccharides anti liver fibrosis, also to lay a certain foundation for the further development of the new liver fibrosis drugs.Method:Randomly selected 5 male SD rats and 5 female SD rats from 70 SD rats (half male, half female) as the normal group(Group A), gave them normal saline injection,and the rest of rats received 40% CCL4 subcutaneous injection and 30% ethanol lavage to construct the model of hepatic fibrosis in rats. After confirmed the formation of liver fibrosis by the pathological slices, randomly divided the remaining 44 rats into 6 groups:Group B, Group C, Group D, Group E, Group F. Then gave the rats the medicine by gavage. Group A and Group B were fed with 1 ml per 100g body weight normal saline;Group C, D were fed with 0.5g per 100g body weight, 1g per 100g body weight dendrobium polysaccharides;Group E were fed with 0.2 mg per 100g body weight colchicine. Group F were fed with 330 mg per 100g body weight fuzheng huayu capsule. The treatment lasted for 4 weeks, after that,kill all the rats, detect the ALT, AST, HA by the blood. The expression of collagen type I, collagen type ?, TIMP-1, MMP-13, TGF-?1 in hepatic tissue was detected by real time quantitative RT-PCR.Results:1.Compared with Group A,the level of ALT,AST, HA in Group B were significantly increased(P<0.05);Compared with Group B, the level of ALT, AST, HA in group C, D, E, F were dereased(P<0.05).2. H&E staining:Compares with Group A, the stage of hepatic fibrosis was apparently increased in group B. Compared with Group B, the stage of hepatic fibrosis in Group C and Group D and group E and group F were decreased.3. Compared with group A, the expression of TIMP-1 in group B increased (P<0.05). Compared with group B, the expression of TIMP-1 in group C, D, E, F decreased (P<0.05). Compared with group A, the expression of TIMP-1 in group B decreased (P<0.05). Compared with group B, the expression of TIMP-1 in group C, D, E, F increased (P<0.05).Conclusions:1. Dendrobium polysaccharides can restore the damage of fibrosis in liver tissue, it shows dendrobium polysaccharides can protect liver fibrosis rats and treat liver fibrosis.2. Dendrobium polysaccharides can reduce the expression of TIMP-1 in liver fibrosis rats, make the expression of MMP-13 raise. It shows dendrobium polysaccharides treat hepatic fibrosis though regulate TIMPs/MMPs, reduce the expression of extracellular matrix in liver and balance liver collogen.
