| With the continuous development and progress of national economy,China is gradually entering an aging society,as a common disease of the older men-benign prostatic hyperplasia(benign prostatic hyperplasia BPH)is a kind of common diseases and frequent diseases in older men,The clinical symptoms of BPH to have serious influence on the quality life and health of old men.Although the studies and reports is riched in scholars both at home and abroad,Western medicine showed certain effect in the treatment of BPH,but its side effects with a long cycle,expensive,and other factors,its treatment effect is still unsatisfactory.Traditional Chinese medicine for the treatment of the disease not only thinking more broad.My tutor,professor Mr Chen engaged in clinical and scientific research and teaching of traditional Chinese medicine for many years,On the basis of the theory of traditional Chinese medicine treatment of male diseases BPH has accumulated rich clinical experience,creating hospital preparations Yishen Tonglong capsule,which is better in applied to the clinical?But the mechanism of BPH is not clear for Yishen Tonglong capsule,now proposed related animal experiment,to explore the effects on reproductive sex hormone in Yishen Tonglong capsule summed up the treatment of benign prostatic hyperplasia treated from kidney,the proposed "kidney tong Long capsule" in the treatment of BPH.Obtain good clinical effect,worthy of further promotion and research.Purpose: To study the effect and the action mechanism of Yishen Tonglong Capsule(YTC)on sex hormone lever in the mouse model of benign prostate hyperplasia?Methods: The BPH model was made in male mice by subcutaneous injection of testosterone propionate at 5mg/kg·d for 3 weeks,after causing modeling success of BPH divided the mice into six groups: normal control group,model group,Longbishu capsules(LBS),and high-,medium-and low-dose YTC group,10 mice in each?Respectively were fed with 1.2,0.6,0.3g/kg YTC suspension,0.45g/kg LBSsuspension,The normal control group and model group were given the same volume of distilled water?After 8 weeks,the mice were sacrificed and their prostate taken for measurement of their wet weight,calculation of the prostatic index(PI),determination Of serum testosterone(T)? estradiol(E2)and E2/T ratio,and observeation the morphological changes at the prostate tissue under the light microscope?Results:(1)General condition of mice:Compared with the normal group,the model group mice Likes to sleep cluster,less activity,hair Shaggy and dry,weight increased?After feed medicine in groups of mice,the behavior of mice have ease,mental state,activity is flexible,increase appetite,hair become moist?(2)Prostate tissue observation: Compared with the normal group,the model group mice prostate tissue increased significantly,glands color is reddish /gray,touch slightly hard,surface is not smooth? In addition to low-dose YTC,compared with model group ignorance changes,the prostate tissue form all have different degrees of improvement in medium-and high-dose YTC groups and LBS group?(3)The wet weight of prostate and PI :The wet weight of prostate and PI were significantly decreased in medium-and high-dose YTC groups and LBS group as compared with the model group(P<0.01)?(4)The serum testosterone(T)?estradiol(E2)and E2/T ratio in different groups of mice : normal control group :(1.73±0.015)pg/ml,(73.08±1.027)pg/ml,(42.30±0.525)pg/ml;model group:(3.86±0.020)pg/ml,(145.79±0.876)pg/ml,(37.76±0.254)pg/ml;LBS group :(2.47±0.018)pg/ml,(95.87±0.472)pg/ml,(38.80±0.134)pg/ml;high-dose YTC:(2.75±0.029)pg/ml,(107.11±0.611)pg/ml,(38.92±0.355)pg/ml;medium-dose YTC:(2.77±0.024)pg/ml,(112.16±0.824)pg/ml,(40.56±0.293)pg/ml;low-dose YTC:2.91±0.026)pg/ml,(112.68±0.774)pg/ml,(38.80±0.424)pg/ml;Compared with the model group,the former with statistically significant dilference(P<0.05 ? P<0.01)?Conclusion: Yishen Tonglong can reduce the levels of T and E2,improve the ratio of E2/T,which manifested the action mechanism of Yishen Tonglong Capsule in the treatment of BPH. |
