| Objective: the experiment is designed to clear Notch signaling pathway and the relationship of angiogenesis in burn wound healing and to observe different doses of nano vanadium ice milk ligand expression of Notch signal pathway and the effect of preliminary discussion alum ice the mechanism of action of microemulsions,promote healing of burn wounds.Methods:(1)120 male and female both New Zealand white rabbit,were randomly divided into blockers group,model group and normal control group.Blockers group and model group to build the day before membrane press 2 mg/kg respectively injection of gamma secretase inhibitors(DAPT)and normal saline,building 1 times a day after the last 14 d,and normal saline groups will not be made mold,the same amount of saline injection for 14 consecutive days.Each group respectively in 3,7,14,21,28 days randomly selected eight executed,using immunohistochemical detection of the expression of VEGF and VEGFR-2.(2)200 male and female both healthy New Zealand white rabbit,randomly divided into normal saline group,hot oil boils,alum ice microemulsions,low,medium and high dose group 5,respectively corresponding experimental drug daub after building,2 times/day,0.2 ml/second,continuous administration 28 days,respectively,in 3,7,14,21,28 days randomly 8 only after death,the Western-blot test Notch ligand(Jagged1,Jagged2 1,Dll,the Dll--3,the Dll-4)protein expression.Results:(1)compared with model group,blockers group 5 time points of dynamic wound healing rate were lower than that of model group,the differences between the two groups have statistical significance(p < 0.01)significantly.(2)compared with normal control group,model group and blockers of each point in time the expression of VEGF and VEGFR-2 difference had statistical significance(p < 0.05);Group compared with model group,blockers of each point in time the expression of VEGF and VEGFR-2 are lower than the expression of model group,the differences between the two groups have statistical significance(p < 0.05).(3)compared with normal saline group,the other four groups in 5 time points of Notch ligand(Jagged1,Dll,Jagged2,Dll-1-3)enhancement of expression to have statistical significance(p < 0.05),especially for alum ice was most evident in the nano emulsion dosage group(p < 0.05),while alum ice microemulsions,low and high dose group was higher than with hot oil the ligand expression differences between groups had no statistical significance(p > 0.05);(4)compared with normal saline group,the other four groups in 5 time points of Notch ligand Dll-4 weakening of expression to have statistical significance(p < 0.05),among them with alum ice dose group was most pronounced in nano emulsion,statistically significant difference(p < 0.05),while alum ice microemulsions,low and high dose group was higher than with hot oil group between the ligand expression had no statistical significance(p > 0.05).Conclusions:(1)In burn wound healing,the Notch signaling pathway blockade is weakened wound angiogenesis in the expression of VEGF and VEGFR-2,decrease the rate of dynamic healing wounds,Notch signal pathway may by intervening in burn wound healing in the expression of vascular active factor to affect the healing of burn wounds(2)the alum ice nano can increase Notch ligands Jagged1,Jagged2 1,Dll,the Dll-cut Dll-4-3 expression and expression,of which the dose group of the most obvious,prompt alum ice the mechanism of action of microemulsions,promote healing of burn wounds to increase Notch ligands Jagged1,Jagged2 1,Dll,the Dll-cut Dll-4-3 expression and the expression of related. |
