Purpose : To examine the effects of Astragalus, Pueraria and the compatibility of Astragalus and Pueraria on factors related the insulin signaling pathway and adiponectin signaling pathway and inflammatory cytokines in pancreas of diabetic rats, aiming to analyze the mechanism of action of Astragalus, Pueraria and the compatibility of Astragalus and Pueraria on pancreas glucose and lipid metabolism in diabetic rats.Material and method:The 86 healthy male SD rats, were randomized to 1 of 6 groups according to blood glucose, which namely control group, model group, positive control(Jinqi metformin) group, Astragalus group, Pueraria group, Astragalus and Pueraria group.There were 15 rats in each group,except control group(11 rats). In addition to the rats in control group, the rats in rest groups were injected intraperitoneally with streptozotocin(46mg?kg-1) to make after diabetes mellitus model. Except the control group and positive control group, the rest groups were implemented experiment according to 2×2 factorial design.Treated with drugs for 30 days,the levels of fasting blood glucose,serum cholesterol and triglyceride wrer assayed. the levels of pancreatic insulin(INS), adiponectin(ADPN),glucose transporter 4(GLUT4), interleukin-12(IL-12),interleukin-15(IL-15)and tumor necrosis factor-?(TNF?) were detected by enzyme-linked immunosorbent assay(ELISA); the expression levels of adiponectin receptor 1(Adipo R1) and AMP-activated protein kinase(AMPK) m RNA in pancreas were assayed by Quantitative Real-Time PCR(QPCR). SPSS software for Windows.version 17.0 were used for performing statistical analyses.Results:1. In model rats, the level of blood glucose increased significantly.Compared with the model group, there were lower level of blood glucose in Astragalus group, Pueraria group,Astragalus and Pueraria group, and the differences were statistically significant(P<0.05).Compatibility of Astragalus and Pueraria did not show interaction effects on blood glucose.2. In model rats, the levels of serum CHO and TG increased significantly.Compared with the model group, there were lower level of serum CHO and TG in Astragalus group, Puerariagroup, Astragalus and Pueraria group, and the differences were statistically significant(P<0.05). Interaction effect analysis:(1)the level of serum CHO: Compatibility of Astragalus and Pueraria did not show interaction effects(P>0.05).(2)the level of serum TG: Compatibility of Astragalus and Pueraria shew interaction effect on the level of serum TG,and the combined was better than any single effect.3. In model rats, the levels of pancreatic INS, ADPN and GLUT4 decreased significantly.(1)the level pancreatic INS: Compared with the model group,there was higher level of INS in Astragalus group, Astragalus and Pueraria group,and there was not significant change in Pueraria group.Compatibility of Astragalus and Pueraria did not show interaction effects.(2)the level of pancreatic ADPN: Compared with the model group,there was higher level of ADPN in Astragalus group, Astragalus and Pueraria group(P < 0.05),and there was not significant change in Pueraria group.Compatibility of Astragalus and Pueraria did not show interaction effects.(4)the level of pancreatic GLUT4: Compared with the model group, there were higher levels of GLUT4 in Astragalus group, Pueraria group, Astragalus and Pueraria group, and the differences were statistically significant(P < 0.05). Compatibility of Astragalus and Pueraria did not show interaction effect.4. In model rats, the levels of pancreatic IL-12,IL-15 and TNF? increased significantly.(1)the level of pancreatic IL-12:Compared with the model group,there were lower levels of pancreatic IL-12 in Astragalus group, Pueraria group, Astragalus and Pueraria group, and the differences were statistically significant(P<0.05). Compatibility of Astragalus and Pueraria shew interaction effect on the level of pancreatic IL-12,and Astragalus' single effect was better than the combined.(2)the level of pancreatic IL-15:Compared with the model group,there were lower levels of pancreatic IL-15 in Astragalus group, Pueraria group, Astragalus and Pueraria group, and the differences were statistically significant(P<0.05). Compatibility of Astragalus and Pueraria shew interaction effect on the level of pancreatic IL-15,and Astragalus' single effect was better than the combined.( 3) the level of pancreatic TNF?:Compared with the model group, there were lower levels of pancreatic TNF? in Astragalus group, Pueraria group, Astragalus and Pueraria group, and the differences were statistically significant(P<0.05). Compatibility of Astragalus and Pueraria shew interactioneffect on the level of pancreatic TNF?,and Astragalus' single effect was better than the combined.5. In model rats, the expression of pancreatic IRS2,PI3 K and Akt2 m RNA decreased significantly.( 1) the IRS2 m RNA:Compared with the model group, there were more expression of pancreatic IRS2 m RNA in Astragalus group, Pueraria group, Astragalus and Pueraria group. Compatibility of Astragalus and Pueraria shew interaction effect on the expression of pancreatic IRS2 m RNA,and Astragalus' single effect was better than the combined.(2)the PI3 K m RNA:Compared with the model group,there were more expression of pancreatic PI3 K m RNA in Astragalus group, Pueraria group, Astragalus and Pueraria group. Compatibility of Astragalus and Pueraria did not show interaction effect on the expression of pancreatic PI3 K m RNA.(3)the Akt2 m RNA:Compared with the model group,there were more expression of pancreatic Akt 2m RNA in Astragalus group, Pueraria group,Astragalus and Pueraria group. Compatibility of Astragalus and Pueraria shew interaction effect on the expression of pancreatic Akt 2m RNA,and Astragalus' single effect was better than the combined.6. In model rats, the expression of pancreatic Adipo R1 and AMPKm RNA decreased significantly.(1)the Adipo R1 m RNA:Compared with the model group,there were more expression of pancreatic Adipo R1 m RNA in Astragalus group, Pueraria group, Astragalus and Pueraria group. Compatibility of Astragalus and Pueraria shew interaction effect on the expression of pancreatic Adipo R1 m RNA,and the single effect was better than the combined.( 2) the AMPKm RNA:Compared with the model group, there were more expression of pancreatic AMPKm RNA in Astragalus group, Pueraria group, Astragalus and Pueraria group.Compatibility of Astragalus and Pueraria shew interaction effect on the expression of pancreatic AMPKm RNA,and the combined single effect was better than any single.Conclusion:1. Astragalus and Pueraria could reduce the levels of blood glucose and serum lipid.The combined effect on reducing the levels of serum TG was better than any single.2. The effect of compatibility of Astragalus and Pueraria on coregulating the blood glucose and serum lipid may be related with regulating the inflammatory cytokines and the IRS2/PI3K/Akt2 pathway stimulated by INS and the Adipo R1/AMPK pathway stimulated byADPN.3. Astragalus and Pueraria emphasized differently on regulating the DM rat pancreas glucolipid metabolism related targets,and the Astragalus' effect was better than Pueraria's and the combined.
