Purpose:To investigate the effect of Angelica Sinensis Polysaccharide(ASP) and Astragalus Polyacharin(APS),on the proliferation of mice bone marrow hematopoietic stem cells(HSCs).To investigate the production of cytokines and the expression level of transcription factors. Then proviod the basis for new drug development.Material and method:Animals were randomly divided into 6 groups, which were normal group(N), model group(M), EPO+G-CSF group, ASP group, APS group and ASP+APS group.Mouse received with either Cyclophosphamide(380 mg·kg-1·d-1,model group) or the equal volume of saline(normal group) daily via peritoneal injection(i.p.) for 3 days. Then the animals were killed,whose BMNCs were cultured at different concentrations containing drug of EPO(50 U·m L-1)+G-CSF(250 U·m L-1),ASP(0.2 g·L-1),APS(0.2 g·L-1)and ASP+APS for four days. MTT method was used to observe the effects of various culture fluid on proliferation of BMNCs,ELISA method was used to observe the production of cytokines EPO?GM-CSF?IL-3?SCF?INF-? and TNF-?,QPCR method was used to observe the expression level of transcription factors GATA-1 and PU.1. Data are processed by SPSS 17.0, expressed as a`x±s, p < 0.05, with statistical significance.Results:1.After be cultured at concentrations, the CFU-E could be seen in normal group,which yielding a colony with not fewer than 8 cells,but these cells could be smaller or non-hemoglobinized.And the fewer CFU-GM could be seen.After the stimulation of various culture fluids, the results in all drug groups suggested BMNCs showed a proliferative property manifesting a higher cell number than model group,and ASP+APS group was better than others.2.The production of cytokines EPO?GM-CSF?IL-3?SCF?INF-? and TNF-? in each group was higher than model group.The production of cytokines EPO?GM-CSF?SCF?IL-3 theASP+APS group was the similar as the EPO+G-CSF group, but was higher than the others.The production of cytokines EPO?GM-CSF and SCF,ASP group was higher than APS group.But the production of cytokine IL-3, the two groups were the same.3.The expression level of transcription factors GATA-1 and PU.1,each group was higher than model group,the ASP+APS group was the similar as the EPO+G-CSF group, but was higher than others.And ASP group was higher than APS group.Conclusion:1.The drugs ASP and APS promotes the proliferation of mice bone marrow hematopoietic stem cell,and the proliferation of mice bone marrow hematopoietic stem cell in the groups ASP and APS was higher than the groups of ASP or APS.2.The drugs ASP and APS promotes the production of cytokines EPO?GM-CSF?IL-3?SCF,and inhibits the production of cytokines INF-? and TNF-?.The culture fluid with drugs ASP and APS was the most effective.3.According to promoting the expression level of hematopoietic transcription factors GATA-1and PU.1,The drugs ASP and APS promotes the proliferation of mice bone marrow hematopoietic stem cell.
