Objective:Worldwide, the HPV prevalence varies among different places and so far, still there is no clear and accurate data about the HPV prevalence of Inner Mongolia, Tongliao district. The intensity of Mongolian residence is up to 46%, in this area and this area is one of the high-prevalence areas of cervical cancer in China. Cervical cancer is caused by HPVs, which is verified by many researches. Therefore, HPVs may be prevalent in this a rea. Meanwhile, there are many researches indicating that those high-risk HPVs have tight relations with the happening of head and neck cancer so we deduce that the local head and neck cancers may have a stong relati on with the local prevalent HPVs. As a consequence, we try to reveal the prevalence and types of HPVs in head and neck cancer in Inner Mongoli a, Tongliao district, by some experimental measures. On one hand, we can initially fill the knowledge gap through this research field. On the other hand, we can provide some references for making strategies aginst head a nd neck cancer, in China.Methods:1. The cases with routine diagnosis of cancer in head and neck area were searched from pathology record of 2 hospitals in Tong-Liao area, the Inner Mongolia University for Nationalities Affiliated Hospital and The Fi rst People's Hospital of Khorchin, Tong-Liao City between October 2009 a nd December 2012.2. The blocks of paraffin-embedded tissues of head and neck cancers obtained were sectioned and those pieces include the initial pieces a (HE pieces), the backup vacant pieces, the PCR pieces, the backup PCR pieces, and the final pieces b (HE pieces).3. Diagnose those cases and input the final outcomes into the identica 1 CRF sheets.4. Extract DNA from those paraffin-embedded tissues with protease K and amplificate the DNA with ?-globin primers(forward 5'-CAACTTCAT CCACGTTCACC-3'and reverse 5'-GAAGAGCCAAGGACAGGTAC-3')5. All specimens were tested by consensus primers nested PCR assay and HPV E6 gene Type-Specific Primers nested PCR for HPV prevalence and types.6. Reextract those unsuccessful extracted specimens by using purifying DNA kit.7. Sequence those HPV positive specimens, and analyse the HPV typ e distribution and the mutation of E6 gene.8. Immunostain those backup vacant pieces for p16INK4a. The positiv e one from cervical cancer as the positive control and observe the p16IN K4a status of our specimens.Results:1. We collected 137 head and neck cancer cases from local hospitals.2. After 2 pathologists' diagoses, we got the accurate diagnostic outco mes of our 137 cases:HNSCCs 47, cancers 7, atypical hyperplasia 8, pap illoma 50, polypus 25.3. In 137 cases,7 HPV positive specimens were detected. In 54 real head and neck cancers,4 HPV positive specimens were detected and 3 HPV positive specimens were detected from 47 HNSCCs.4. Outcomes of sequencing:case 7th:HPV 58, case 33rd and 41st:H PV 16, case 68th:HPV 31, case 75th:HPV 81, case 93rd:HPV 6, case 99th:HPV 11, and cases 7,33,41 are HNSCC.5. The p16INK4a positive rate of 47 HNSCCs is 10.6% and thep16I NK4a positive rate of other cancers is 42.9%. The overall p16INK4a pos itive rate of head and neck cancer is 16.7%.Conclusion:1. The HPV prevalence among head and neck cancer in this area is 7.4%. The HPV prevalence among HNSCCs in this area is 6.4%. The HPV prevalences of two researches of Beijing are 27.4% and 17.2% respectively and The HPV prevalence of one research in Shanghai is 3.5%. This conclusion demonstrate the differentiation of HPV prevalence among different regions in China.2. The potential oncogenic HPV types for head and neck cancer in Inner Mongolia, Tongliao district are HPV 58, HPV 16, HPV 81, HPV 6, HPV 11, HPV 31. The potential oncogenic HPV types for HNSCC in this area are HPV 58 and HPV 16 and they are high-risk HPVs...
