Purpose:To discuss the molecular biological mechanisms of SHENSHUAIYIN restrain renal fibrosis by molecular biology, immunohistochemistry methods.Provide animal experiment basis for effective treatment of traditional Chinese medicine of chronic renal failure. Material and method:Using the kidney disease model made by adenine gavage method,60 healthy male SD rats(body weight 180 ± 20)g were randomly extracted with 12 normal group, the others are the model group,use adenine per 200 mg /(kg·d) doses to gavage rats,once a day,the normal group fed with normal volume 8ml0.9%NaCl day gavage for 3 weeks.When the fourth weeks,all rats were used to make membrane model using randomly subdivided into SHENSHUAIYIN group, model group and Uremic clearance group.SHENSHUAIYIN group by crude drug 33.3g /(kg·d) dose gavage renal failure decoction;Uremic clearance group by Uremic clearance granules 2.25 g /(kg·d) dose gavage; model group and the normal group with the same group dose gavage 0.9% NaCl solution.In the experiment seven weeks later,all of these rats were unified with 10% chloral hydrate intraperitoneal injection of anesthesia, blood taken from the abdominal aorta was sacrificed and the kidneys were taken.RT-PCR method was used to detect the effect of renal failure on the expression of TGF-?1mRNA in CRF rats.ELISA method was used to detect the expression of Smad2, Smad3 and Smad7 protein, and the expression of ?-SMA protein was detected by immunohistochemistry.The results were analysed by the analysis of variance in SPSS17.0 software. Results: 1.TGF-?1 mRNA expression in model group increased, there was a significant difference(P<0.05) with the normal group; compared with the model group, SHENSHUAIYIN group and Uremic clearance group decreased(P<0.01), Compared with Uremic clearance group, SHENSHUAIYIN group of TGF-?1mRNA expression decreased,there was a significant difference(P<0.01). 2. Compared with the normal group, the levels of Smad2, Smad3 of Model group, SHENSHIYIN group and Uremic clearance group increased(P<0.01), Smad7 decreased(P<0.05), after treatment, the levels of Smad2, Smad3 of SHENSHUAIYIN group and Uremic clearance group decreased(P<0.01), Smad7 increased(P<0.01), but the result of SHENSHUAIYIN group better than Uremic clearance group group(P<0.05). 3.Compared with the normal group, the express of ?-SMA protein of model group, SHENSHIYIN group and Uremic clearance group significantly increased(P<0.01);compared with the model group,SHENSHUAIYIN group and Uremic clearance group decreased(P<0.05);compared with Uremic clearance group,the express of ?-SMA protein were decreased in SHENSHUAIYIN group(P<0.05). Conclusion: 1. SHENSHIYIN can inhibit TGF-?1mRNA expression in renal fibrosis, delaying the progression of renal fibrosis. 2. SHENSHIYIN can inhibit Smad2, Smad3 levels and increase the level of Smad7 by regulating TGF-?1 / Smads signaling pathway to inhibit the occurrence and development of renal fibrosis. 3.SHENSHIYIN can significantly repair rats' kidney tissue damaged by adenine, thus confirming the effectiveness of the treatment of chronic renal failure, renal failure drink.
